摘要:【目的】探究地衣芽孢桿菌(Bacillus licheniformis)對(duì)草魚(Ctenopharyngodon idellus)生長(zhǎng)性能、肌肉品質(zhì)及土腥味防控的影響,為草魚養(yǎng)殖中土腥味防控與肌肉品質(zhì)提升提供參考依據(jù)?!痉椒ā窟x取360尾(體質(zhì)量25.00±1.50 g)草魚,隨機(jī)分為T1(投喂基礎(chǔ)飼料)、T2(投喂地衣芽孢桿菌飼料)、T3[投喂基礎(chǔ)飼料并添加嗜熱一氧化碳鏈霉菌(Streptomyces thermocarboxydus)和藍(lán)微褐鏈霉菌(S.cyaneofuscatus)]、T4(投喂地衣芽孢桿菌飼料并添加嗜熱一氧化碳鏈霉菌和藍(lán)微褐鏈霉菌)組。飼喂30 d,檢測(cè)地衣芽孢桿菌對(duì)草魚生長(zhǎng)性能、肌肉質(zhì)構(gòu)特性、肌肉常規(guī)營(yíng)養(yǎng)成分含量、肌肉脂肪酸含量及土腥味物質(zhì)濃度的影響?!窘Y(jié)果】飼料中添加地衣芽孢桿菌能有效提升草魚生長(zhǎng)性能并改善肌肉質(zhì)構(gòu)特性,與T1組相比,T2組草魚增重率和特定生長(zhǎng)率顯著升高(Plt;0.05,下同),T2和T4組草魚肌肉硬度顯著大于T1和T3組。飼料中添加地衣芽孢桿菌能有效提升草魚肌肉品質(zhì),T2和T4組草魚肌肉粗蛋白含量和多不飽和脂肪酸總含量顯著高于T1和T3組,二十二碳六烯酸、二十碳五烯酸和花生二烯酸顯著高于T3組。飼料中添加地衣芽孢桿菌能有效降低草魚肌肉中土腥味物質(zhì)濃度,T2組草魚肌肉土臭素濃度顯著低于T1、T3和T4組,T3組草魚肌肉中2-甲基異莰醇濃度顯著高于T1、T2和T4組。相關(guān)分析結(jié)果表明,土臭素和2-甲基異莰醇濃度均與多不飽和脂肪酸總含量呈負(fù)相關(guān),土臭素濃度與粗脂肪含量呈顯著正相關(guān)?!窘Y(jié)論】飼料中添加地衣芽孢桿菌能改善草魚的生長(zhǎng)性能,優(yōu)化肌肉營(yíng)養(yǎng)組成,提升肌肉質(zhì)構(gòu)特性并降低肌肉中土腥味物質(zhì)積累。地衣芽孢桿菌可能是通過調(diào)控脂肪酸代謝,以減少草魚肌肉中土腥味物質(zhì)積累。

Effects of Bacillus licheniformis on the growth performance, muscle quality and earthy odor control of grass carp (Ctenopharyngodonidellus)

Abstract:【Objective】To investigate the effects of Bacillus licheniformis on the growth performance,muscle quality and earthy odor control of grass carp(Ctenopharyngodon idellus),which could provide reference for the prevention and control of earthy odor in cultured C.idellus and the improvement of muscle quality.【Method】A total of 360 C.idellus(with an initial weight of 25.00±1.50 g)were randomly divided into 4 groups:T1(fed with basic diet),T2(fed with B.licheniformis diet),T3(fed with basic diet adding Streptomyces thermocarboxydusand S.cyaneofuscatus),T4(fed with B.licheniformis diet adding S.thermocarboxydus and S.cyaneofuscatus).After a 30 d feeding trial,the effects of B.li-cheniformis on grass carp growth performance,muscle texture characteristics,muscle conventional nutrient content,muscle fatty acid content and earthy odor substance concentration were detected.【Result】The addition of B.licheniformis could effectively improve the growth performance and muscle texture characteristics of C.idellus.Compared with the T1 group,the weight gain rate and specific growth rate of the T2 group were significantly increased(Plt;0.05,the same be-low).The muscle hardness of the T2 and T4 groups was significantly greater than that of the T1 and T3 groups.The addi-tion of B.licheniformis to feed could effectively enhance the muscle quality of C.idellus.The crude protein content and to-tal polyunsaturated fatty acid content in the muscles of the T2 and T4 groups were significantly higher than those of the T1 and T3 groups respectively,and the contents of docosahexaenoic acid,eicosapentaenoic acid and arachidonic acid were all significantly higher than those of the T3 group.The addition of B.licheniformis to feed could effectively reduce the earthy odor substance concentration in C.idellus muscle,with group T2 showing significantly lower geosmin(GSM)concentration than T1,T3 and T4 groups.The 2-methylisoborneol(2-MIB)concentration in group T3 was significantly higher than in T1,T2 and T4 groups.Correlation analysis results indicated that the concentrations of GSM and 2-MIB were both negatively correlated with the total content of polyunsaturated fatty acids,and the concentration of GSM wassignificantly and positively correlated with the crude fat content.【Conclusion】Adding B.licheniformis to the basic diet can enhance the growth performance of C.idellus,optimize muscle nutritional composition,improve texture characteris-tics and reduce earthy odor substance accumulation in muscle.B.licheniformis may reduce the accumulation of earthy odor substance in C.idellus muscle by regulating fatty acid metabolism.

0引言
【研究意義】草魚(Ctenopharyngodon idellus)是我國(guó)主要的養(yǎng)殖魚種之一,2023年全國(guó)草魚產(chǎn)量超過594萬t,位居我國(guó)魚類產(chǎn)量首位(農(nóng)業(yè)農(nóng)村部漁業(yè)漁政管理局等,2024)。草魚養(yǎng)殖過程中常面臨土腥味問題,影響草魚的品質(zhì)和市場(chǎng)價(jià)值(劉利平等,2021)。目前,常用流水吊養(yǎng)方式減輕魚體土腥味,但該方法存在暫養(yǎng)周期長(zhǎng)和流水用量大等問題,且只能減輕部分土腥味,并推遲了上市時(shí)期,無法從根本上解決草魚土腥味和品質(zhì)下降問題。因此,探究解決草魚土腥味的技術(shù)和方法,在養(yǎng)殖過程中提升草魚品質(zhì),對(duì)促進(jìn)草魚養(yǎng)殖業(yè)健康可持續(xù)發(fā)展具有重要意義?!厩叭搜芯窟M(jìn)展】土臭素(Geosmin,GSM)和2-甲基異莰醇(2-methylisoborneol,2-MIB)是構(gòu)成土腥味物質(zhì)的主要成分,二者共同導(dǎo)致水體和魚體產(chǎn)生土腥味(Howgate,2004),嚴(yán)重影響草魚品質(zhì)。在水產(chǎn)養(yǎng)殖中,土腥味物質(zhì)主要由微生物和微藻代謝產(chǎn)生(Schrader etal.,2010)。這些微生物在富含營(yíng)養(yǎng)元素的水體中大量繁殖,特別是在高溫和低氧等環(huán)境中,其代謝活性增強(qiáng),會(huì)產(chǎn)生更多土腥味物質(zhì)(劉利平等,2021)。此外,池塘底泥中有機(jī)物的腐敗分解、過高的養(yǎng)殖密度、飼料使用不當(dāng)及水體流動(dòng)不暢也會(huì)加劇土腥味問題(閆莉等,2022)。因此,如何降低草魚土腥味、提升草魚魚肉品質(zhì)成為當(dāng)前亟待解決的問題。鏈霉菌是產(chǎn)生土腥味物質(zhì)的主要來源,廣泛分布于土壤和水環(huán)境中,其中嗜熱一氧化碳鏈霉菌(Streptomyces thermocarboxydus)和藍(lán)微褐鏈霉菌(S.cyaneofuscatus)在水產(chǎn)養(yǎng)殖中最常見(Gutt-man and Rijn,2008)。張凱等(2018)研究發(fā)現(xiàn),采用底排污—魚菜共生組合模式養(yǎng)殖羅非魚(Oreochro-mis niloticus),能降低其肌肉中的土腥味物質(zhì)濃度,該養(yǎng)殖模式通過改善水質(zhì)和底層環(huán)境,減少了土腥味物質(zhì)的生成和積累,提升了羅非魚的肉質(zhì)和口感;Schram等(2021)研究發(fā)現(xiàn),增大養(yǎng)殖池水體的交換率能減少大西洋鮭(Salmo salar)魚肉中土腥味物質(zhì)的積累;魯強(qiáng)等(2023)通過在清水池塘中進(jìn)行暫養(yǎng)處理,顯著減少了大口黑鱸(Micropterus salmoides)魚肉中GSM含量。地衣芽孢桿菌(Bacillus licheni-formis)在水產(chǎn)養(yǎng)殖中被廣泛用于調(diào)節(jié)水質(zhì)(張澤龍等,2022),同時(shí)還具有促進(jìn)魚類消化和生長(zhǎng)等作用(徐亞飛,2018;張冬梅等,2022),其中地衣芽孢桿菌(B.licheniformis)能通過調(diào)節(jié)腸道菌群組成,促進(jìn)有益菌的生長(zhǎng)、抑制病原菌的繁殖,而改善魚體腸道健康并增強(qiáng)其消化吸收能力(徐亞飛,2018;張宇柔等,2022)?!颈狙芯壳腥朦c(diǎn)】目前,關(guān)于地衣芽孢桿菌的研究多集中于魚類免疫與抗病等方面(曹煜成等,2013;樊英等,2021;王旭達(dá)等,2021),而其對(duì)魚類生長(zhǎng)性能、肌肉品質(zhì)及土腥味的影響鮮見報(bào)道。【擬解決的關(guān)鍵問題】針對(duì)草魚養(yǎng)殖中的土腥味問題,通過拌餌投喂方式,探究地衣芽孢桿菌對(duì)草魚生長(zhǎng)性能、肌肉品質(zhì)及土腥味物質(zhì)濃度的影響,為提升草魚養(yǎng)殖效率和改善魚肉品質(zhì)提供參考依據(jù)。
1材料與方法
1.1試驗(yàn)材料
1.1.1供試菌株嗜熱一氧化碳鏈霉菌菌株(BNCC 153409)和藍(lán)微褐鏈霉菌菌株(BNCC 152414)購(gòu)自北納生物,地衣芽孢桿菌菌株(LL1)由上海海洋大學(xué)水產(chǎn)與生命學(xué)院魚類繁殖與生態(tài)養(yǎng)殖實(shí)驗(yàn)室分離培養(yǎng)所得。
1.1.2供試草魚及飼料供試草魚幼魚購(gòu)自浙江省湖州市草魚養(yǎng)殖場(chǎng),體表無傷、狀態(tài)良好。運(yùn)回實(shí)驗(yàn)室暫養(yǎng)7 d,養(yǎng)殖水體24h供氧(溶氧gt;5 mg/L),水溫(27±2)℃,pH 7.5~8.0。暫養(yǎng)期間,于每日8:00、12:00、16:00、20:00投喂基礎(chǔ)飼料(蛋白含量30%,粒徑2~3 mm)(通威集團(tuán)有限公司),達(dá)飽食水平。
1.2試驗(yàn)設(shè)計(jì)
益生菌飼料制備:將LL1菌株通過平板劃線于37℃恒溫振蕩(175 r/min)培養(yǎng),采用WGZ-XT型智能細(xì)菌濁度儀(上海豐臨電子科技有限公司)測(cè)定活菌數(shù)。將純水稀釋后的菌液均勻噴灑至飼料表面,30℃烘干后于4℃密封保存?zhèn)溆?。飼料中活菌?shù)采用平板菌落計(jì)數(shù)法進(jìn)行測(cè)定,確保其達(dá)到107 CFU/g(王旭達(dá)等,2021)。益生菌飼料每5 d制備1次。
試驗(yàn)設(shè)T1(投喂基礎(chǔ)飼料)、T2(投喂地衣芽孢桿菌飼料)、T3(投喂基礎(chǔ)飼料并添加嗜熱一氧化碳鏈霉菌和藍(lán)微褐鏈霉菌)和T4(投喂地衣芽孢桿菌飼料并添加嗜熱一氧化碳鏈霉菌和藍(lán)微褐鏈霉菌)組,每組設(shè)3個(gè)重復(fù)。試驗(yàn)開始前,將360尾體質(zhì)量25.00±1.50 g、狀態(tài)良好的草魚隨機(jī)分配至12個(gè)循環(huán)水玻璃缸中(60 cm×40 cm×90 cm),每缸30尾。養(yǎng)殖水體中嗜熱一氧化碳鏈霉菌總濃度為106 CFU/mL(嗜熱一氧化碳鏈霉菌與藍(lán)微褐鏈霉菌混合比例為1∶1),各組均添加鏈霉菌碳氮源(紅糖3%、豆粕3%、玉米粉1%、尿素0.25%)。由預(yù)試驗(yàn)結(jié)果可知,飼喂30 d即可使羅非魚幼魚魚肉中富集土腥味物質(zhì)。因此,本試驗(yàn)周期設(shè)為30 d,期間不換水,每日吸污1次,投喂頻率與暫養(yǎng)期間一致。
試驗(yàn)開始前,隨機(jī)選取9尾未分配至循環(huán)水玻璃缸中的草魚,采集背部肌肉用于測(cè)定初始魚肉土腥味物質(zhì)濃度。試驗(yàn)開始時(shí),每缸隨機(jī)選取3尾魚,測(cè)量體質(zhì)量。試驗(yàn)結(jié)束后,禁食24h后采樣,每缸隨機(jī)取3尾魚,低溫麻醉后,測(cè)量體長(zhǎng)和體質(zhì)量;解剖后,測(cè)量?jī)?nèi)臟團(tuán)質(zhì)量;取背部肌肉約6 g,進(jìn)行質(zhì)構(gòu)特性檢測(cè);剩余樣品于-80℃冰箱保存?zhèn)溆?。?dòng)物試驗(yàn)由上海海洋大學(xué)動(dòng)物倫理委員會(huì)批準(zhǔn),批準(zhǔn)號(hào)SHOU-DW-2019-058。
1.3生長(zhǎng)性能指標(biāo)測(cè)定
計(jì)算草魚增重率(Weight gain rate,WGR)、特定生長(zhǎng)率(Specific growth rate,SGR)、存活率(Sur-vival rate,SR)、肥滿度(Condition factor,CF)和臟體比(Visceral somatic indices,VSI)。計(jì)算公式如下:
式中,Wt為草魚終末平均體質(zhì)量(g);W0為草魚初始平均體質(zhì)量(g);t為試驗(yàn)天數(shù)(d);Nf為終末草魚尾數(shù);Ni為初始草魚尾數(shù);L為草魚終末體長(zhǎng)(cm);Wv為草魚終末內(nèi)臟團(tuán)質(zhì)量(g)。
1.4樣品測(cè)定
1.4.1質(zhì)構(gòu)特性測(cè)定參照郭曉東(2019)的方法,進(jìn)行魚肉質(zhì)構(gòu)特性測(cè)定。
1.4.2營(yíng)養(yǎng)成分測(cè)定按照鮮重質(zhì)量計(jì)算各營(yíng)養(yǎng)成分的含量。采用GB 5009.5—2016《食品安全國(guó)家標(biāo)準(zhǔn)食品中蛋白質(zhì)的測(cè)定》中的凱氏定氮法測(cè)定魚肉中粗蛋白含量;采用GB 5009.6—2016《食品安全國(guó)家標(biāo)準(zhǔn)食品中脂肪的測(cè)定》中的索氏抽提法測(cè)定魚肉中粗脂肪含量;采用GB 5009.3—2016《食品安全國(guó)家標(biāo)準(zhǔn)食品中水分的測(cè)定》中的直接干燥法測(cè)定魚肉中水分含量;采用GB 5009.4—2016《食品安全國(guó)家標(biāo)準(zhǔn)食品中灰分的測(cè)定》中的馬弗爐灼燒法測(cè)定魚肉中粗灰分含量;采用GB 5009.168—2016《食品安全國(guó)家標(biāo)準(zhǔn)食品中脂肪酸的測(cè)定》中的內(nèi)標(biāo)法測(cè)定魚肉中脂肪酸種類和含量。
1.4.3土腥味物質(zhì)測(cè)定參考Gharti等(2023)的方法,進(jìn)行魚肉中土腥味物質(zhì)提取前處理及測(cè)定。
1.5統(tǒng)計(jì)分析
試驗(yàn)數(shù)據(jù)采用SPSS 20.0進(jìn)行單因素方差分析(One-way ANOVA),以Duncan’s新復(fù)極差法進(jìn)行多重比較。
2結(jié)果與分析
2.1地衣芽孢桿菌對(duì)草魚生長(zhǎng)性能的影響
地衣芽孢桿菌對(duì)草魚生長(zhǎng)性能的影響見表1。T2組草魚增重率(56.19%)和特定生長(zhǎng)率(1.47%/d)顯著高于T1組(Plt;0.05,下同)。T3組草魚增重率(18.63%)和特定生長(zhǎng)率(0.56%/d)顯著低于T1組。T4組草魚增重率(19.03%)和特定生長(zhǎng)率(0.57%/d)顯著低于T2組。T1和T2組草魚存活率、肥滿度和臟體比均無顯著差異(Pgt;0.05,下同)。
2.2地衣芽孢桿菌對(duì)草魚肌肉質(zhì)構(gòu)特性的影響
地衣芽孢桿菌對(duì)草魚肌肉質(zhì)構(gòu)特性的影響見表2。T2和T4組草魚肌肉硬度顯著大于T1和T3組。T2組草魚肌肉咀嚼性大于T1組,但無顯著差異。T4組草魚肌肉咀嚼性顯著大于T3組。T4組草魚肌肉彈性、黏聚性和膠著性均大于T3組,但差異不顯著。T2組彈性、黏聚性、咀嚼性、回復(fù)性和膠著性均大于T1組,但差異不顯著。
2.3地衣芽孢桿菌對(duì)草魚肌肉常規(guī)營(yíng)養(yǎng)成分含量的影響
地衣芽孢桿菌對(duì)草魚肌肉常規(guī)營(yíng)養(yǎng)成分含量的影響見表3。T2組草魚肌肉粗脂肪含量低于T1組,但無顯著差異。T2和T4組草魚肌肉粗蛋白含量顯著高于T1和T3組。T2和T3組草魚肌肉粗灰分含量顯著低于T1和T4組。T1和T2組草魚肌肉水分含量差異不顯著。
2.4地衣芽孢桿菌對(duì)草魚肌肉脂肪酸含量的影響
草魚肌肉中共檢測(cè)出12種飽和脂肪酸、2種單不飽和脂肪酸和3種多不飽和脂肪酸(表4)。T4組草魚肌肉中二十二碳六烯酸、二十碳五烯酸和花生二烯酸含量均顯著高于T1和T3組。T2組草魚肌肉二十碳五烯酸、二十二碳六烯酸和花生二烯酸含量均顯著高于T3組。T2和T4組草魚肌肉中多不飽和脂肪酸總含量顯著高于T1和T3組。
2.5地衣芽孢桿菌對(duì)草魚肌肉土腥味物質(zhì)濃度的影響
隨著養(yǎng)殖時(shí)間的延長(zhǎng),魚肉會(huì)逐漸富集土腥味物質(zhì),草魚肌肉土腥味物質(zhì)濃度檢測(cè)結(jié)果如圖1所示。T1、T3和T4組草魚肌魚肉中土臭素和2-甲基異莰醇濃度顯著高于Initial組;T2組草魚肌肉中土臭素濃度顯著低于T1、T3和T4組,比T1組低38.46%。T2組草魚肌肉中2-甲基異莰醇濃度顯著低于T1、T3、T4,比T1組低40.91%。T3組草魚肌肉中2-甲基異莰醇濃度顯著高于其他組,比T4組高77.42%。T3組草魚肌肉中的土臭素濃度高于T4組,但差異不顯著。
草魚肌肉土腥味物質(zhì)濃度與臟體比、肥滿度、粗脂肪、多不飽和脂肪酸總含量相關(guān)性分析結(jié)果見表5。草魚肌肉中土臭素和2-甲基異莰醇濃度均與多不飽和脂肪酸總含量呈負(fù)相關(guān),土臭素濃度與粗脂肪含量呈顯著正相關(guān),2-甲基異莰醇濃度與臟體比、肥滿度、粗脂肪含量均無顯著相關(guān)性。
3討論
3.1地衣芽孢桿菌對(duì)草魚生長(zhǎng)性能的影響
研究表明,芽孢桿菌有助于改善魚體腸道健康(Ma et al.,2022),提高飼料吸收率(王麗芳和滿達(dá)虎,2009),促進(jìn)魚類生長(zhǎng)(Ghosh et al.,2002),提高魚體增重率和特定生長(zhǎng)率(沈文英等,2011)。Abarike等(2018)研究表明,飼料中添加地衣芽孢桿菌可顯著提高羅非魚體質(zhì)量、特定生長(zhǎng)率和飼料轉(zhuǎn)化率;王成強(qiáng)等(2019)研究發(fā)現(xiàn),飼料中添加枯草芽孢桿菌(B.subtilis)能明顯促進(jìn)珍珠龍膽石斑魚(Epinephelusfus coguttatus♀×E.slanceolatus♂)幼魚生長(zhǎng)性能。本研究發(fā)現(xiàn),飼料中添加地衣芽孢桿菌的T2組草魚終末平均體質(zhì)量、增重率和特定生長(zhǎng)率均顯著高于其他組。研究表明,地衣芽孢桿菌調(diào)節(jié)消化酶活性(Reda and Selim,2015)的同時(shí)能產(chǎn)生有機(jī)酸和維生素等物質(zhì)(Irianto and Austin,2010),協(xié)助分解腸道中難消化的成分,最終促進(jìn)營(yíng)養(yǎng)物質(zhì)的吸收。提示酶活性的提高和營(yíng)養(yǎng)物質(zhì)吸收能力的加強(qiáng),可能是T2組草魚生長(zhǎng)性能優(yōu)于T1組的原因。此外,T3和T4組草魚生長(zhǎng)性能不理想,可能是受養(yǎng)殖水體中添加的土腥味物質(zhì)嗜熱一氧化碳鏈霉菌的影響。Gagné等(1999)研究發(fā)現(xiàn),虹鱒肝細(xì)胞暴露于0.45 mg/L土臭素48h后,其DNA斷裂顯著增加;Zhou等(2020)研究表明,土臭素可在斑馬魚(Danio rerio)胚胎發(fā)育過程中誘導(dǎo)氧化應(yīng)激、細(xì)胞凋亡和內(nèi)分泌紊亂等多種反應(yīng)。進(jìn)一步說明,T3和T4組草魚生長(zhǎng)性能不理想可能是因?yàn)椴蒴~長(zhǎng)時(shí)間暴露于富含土腥味物質(zhì)的水體環(huán)境中,導(dǎo)致其生理狀態(tài)受影響所致。
3.2地衣芽孢桿菌對(duì)草魚肌肉品質(zhì)的影響
研究表明,水產(chǎn)動(dòng)物體成分影響其肉質(zhì)和口感(柳明等,2010)。周曉波等(2014)研究發(fā)現(xiàn),在奧尼羅非魚(O.niloticus)飼料中添加戊糖乳桿菌(Lacto-bacillus pentosus)后,其魚肉中粗蛋白含量顯著增加,推測(cè)戊糖乳桿菌提高了腸道消化酶活性或補(bǔ)充了額外的營(yíng)養(yǎng)物質(zhì),從而促進(jìn)了魚體蛋白沉積;Adorian等(2018)研究發(fā)現(xiàn),飼喂添加枯草芽孢桿菌和地衣芽孢桿菌飼料的尖吻鱸(Lates calcarifer)肌肉中粗蛋白含量顯著增加,粗脂肪含量顯著減少;趙怡迪(2021)研究表明,復(fù)合益生菌能通過改鯉魚(Cyprinus carpio)腸道消化吸收能力來增加鯉魚肌肉中蛋白含量,進(jìn)而提高鯉魚生長(zhǎng)性能。本研究結(jié)果與上述報(bào)道相符,飼料中添加地衣芽孢桿菌一定程度上提高了草魚肌肉中粗蛋白含量,并降低了粗脂肪含量。提示飼料中添加地衣芽孢桿菌可能改善了草魚消化能力,促進(jìn)了營(yíng)養(yǎng)物質(zhì)的消化吸收,進(jìn)而提高了魚體生長(zhǎng)性能并改善了肌肉營(yíng)養(yǎng)組成。
質(zhì)構(gòu)是食品組織特性的關(guān)鍵指標(biāo)之一(吳朝朝等,2015),也是判斷魚肉新鮮度和感官品質(zhì)的重要標(biāo)準(zhǔn)(劉婧懿等,2020)。胡芬等(2011)研究表明,當(dāng)魚肉具有較高的硬度、彈性和咀嚼性時(shí),其口感通常更佳。Zhou等(2015)研究發(fā)現(xiàn),飼料中添加枯草芽孢桿菌、地衣芽孢桿菌和納豆芽孢桿菌(B.natto)能改善肉雞(Gallus gallus domesticus)肌肉的咀嚼性、持水性和剪切力等質(zhì)構(gòu)屬性;夏雨等(2020)研究發(fā)現(xiàn),飼料中添加乳桿菌(Lactobacillus)能提高凡納濱對(duì)蝦(Litopenaeus vannamei)肌肉彈性和咀嚼性。本研究發(fā)現(xiàn),飼料中添加地衣芽孢桿菌的后草魚肉肌肉硬度顯著提高,表明飼料中添加地衣芽孢桿菌能提升草魚魚肉口感。劉婧懿等(2020)研究發(fā)現(xiàn),魚肉質(zhì)構(gòu)特性主要由肌纖維特性、魚肉基質(zhì)蛋白及魚肉脂肪含量等因素決定,其中魚肉蛋白含量與硬度呈顯著正相關(guān),魚肉中性脂肪含量與硬度呈負(fù)相關(guān)。本研究結(jié)果與之相符,飼料中添加地衣芽孢桿菌提高了魚肉硬度等質(zhì)構(gòu)特性。
脂肪酸是魚肉中重要的營(yíng)養(yǎng)成分,高含量的不飽和脂肪酸可使魚肉更加鮮嫩、口感更好(王萍等,2008)。徐海燕等(2006)研究發(fā)現(xiàn),枯草芽孢桿菌可產(chǎn)生揮發(fā)性脂肪酸;John等(2018)研究證實(shí),飼料中加入枯草芽孢桿菌或熒光假單胞菌(Pseudomonas fluorescens)能提高馬氏沼蝦(Macrobrachium mal-colmsonii)肌肉多不飽和脂肪酸含量。本研究發(fā)現(xiàn),飼料中添加地衣芽孢桿菌的T2和T4組草魚肌肉中多不飽和脂肪酸總含量均顯著高于T1和T3組。Yang等(2010)、Liao等(2015)研究發(fā)現(xiàn),丁酸梭菌可用于改善雞肉脂肪酸組成;Dayangac和Erdem(2017)研究表明,地衣芽孢桿菌可改善消化道消化功能并影響胃腸道中多不飽和脂肪酸的消化與吸收過程,對(duì)動(dòng)物健康存在積極作用。提示地衣芽孢桿菌可能是通過有益代謝物來改善草魚肌肉多不飽和脂肪酸含量,進(jìn)而提升魚肉品質(zhì)。
3.3地衣芽孢桿菌對(duì)草魚肌肉土腥味物質(zhì)的影響
微生物降解法被認(rèn)為是解決土腥味問題最有前景的方法之一,其中最受關(guān)注的是芽孢桿菌的使用(Mcdowall et al.,2009;韓正雙等,2016)。芽孢桿菌作為降解菌,在水環(huán)境中利用2-MIB碳源進(jìn)行生長(zhǎng)并形成孢子,進(jìn)而消除2-MIB(Lauderdale et al.,2004)。Lauderdale等(2004)成功分離出一株可降解2-MIB的芽孢桿菌;馬念念等(2015)研究發(fā)現(xiàn),枯草芽孢桿菌對(duì)GSM和2-MIB降解效率達(dá)90%以上。另外,芽孢桿菌胞外酶的存在使其對(duì)土腥味物質(zhì)具有降解作用。Silvey和Roach(1964)研究發(fā)現(xiàn),表層水中放線菌的減少和土腥味的降低與芽孢桿菌的生長(zhǎng)有關(guān),將土臭素標(biāo)品加入含蠟狀芽孢桿菌的廢棄培養(yǎng)濾液中,81%的GSM被降解。此外,Ho等(2007)認(rèn)為,2-MIB和GSM易受生物降解的原因是其結(jié)構(gòu)中存在可生物降解的脂環(huán)醇和酮,降解過程是一種偽零級(jí)反應(yīng)。本研究發(fā)現(xiàn),飼料中添加地衣芽孢桿菌,GSM和2-MIB濃度均降低,驗(yàn)證了其抑制土腥味的功效。此外,劉利平等(2021)研究發(fā)現(xiàn),水體中添加鏈霉菌后,草魚魚肉GSM含量升高,推測(cè)土腥味物質(zhì)主要通過攝食、鰓呼吸和魚體表皮膚進(jìn)入魚體內(nèi)。Secor和Carey(2016)、Lv等(2018)等研究顯示,魚肉脂肪酸代謝分解過程會(huì)產(chǎn)生游離脂肪酸,導(dǎo)致魚肉中不飽和脂肪酸含量增加,而脂肪醇和脂肪醛含量則會(huì)減少,有助于減少魚肉中異味物質(zhì)的含量,并增加風(fēng)味物質(zhì)的積累。本研究發(fā)現(xiàn),飼料中添加地衣芽孢桿菌,魚肉中土腥味物質(zhì)濃度降低,究其原因可能是草魚肌肉含有較多的多不飽和脂肪酸。
4結(jié)論
飼料中添加地衣芽孢桿菌能改善草魚的生長(zhǎng)性能,優(yōu)化肌肉營(yíng)養(yǎng)組成,提升肌肉質(zhì)構(gòu)特性并降低肌肉中土腥味物質(zhì)積累。地衣芽孢桿菌可能是通過調(diào)控脂肪酸代謝,以減少草魚肌肉中土腥味物質(zhì)積累。
