重樓屬ParisL.植物為藜蘆科(Melanthiaceae)多年生草本植物,生長(zhǎng)在常綠闊葉林、落葉闊葉林、針葉林、竹叢和灌木叢中[1-2];包括33 個(gè)種和15 個(gè)變種[2],主要分布于歐亞大陸的熱帶和溫帶地區(qū),中國大部分地區(qū)均有分布。西南地區(qū)居多[2-3]。重樓屬植物有很高的藥用價(jià)值,在民間,有十幾個(gè)種或變種作為藥用植物被利用。據(jù)《中國藥典》2020 年版記載,重樓藥材的基原植物是云南重樓(滇重樓)ParipolyphyllaSmith var.yunnanensis(Franch.)Hand.-Mazz.和七葉一枝花P.polyphyllaSmith var.chinensis(Franch.) Hara。重樓具有清熱解毒、消腫止痛、涼肝定驚等功效,用于治療疔瘡癰腫、咽喉腫痛、蛇蟲咬傷、跌撲傷痛、驚風(fēng)抽搐等病癥[4]。云南重樓提取物[5]和球藥隔重樓P.fargesiiFranch.根莖提取物[6]已被批準(zhǔn)為化妝品原料。
騰沖重樓P.tengchongensisY.H.Ji, C.J.Yang &Y.L.Huang 僅發(fā)現(xiàn)于云南,生長(zhǎng)于海拔2 750~3 200 m 常綠闊葉林、竹林和灌叢中[7],騰沖重樓被當(dāng)?shù)鼐用癞?dāng)作重樓藥材使用,一般用于治療毒蛇咬傷和跌打傷痛[8]。目前,關(guān)于騰沖重樓化學(xué)成分研究的文獻(xiàn)較少,僅報(bào)道了從騰沖重樓中分離得到的20 個(gè)甾體皂苷類化合物,其中,化合物paristengoside A對(duì)人紅白細(xì)胞白血病HEL 細(xì)胞和人乳腺癌MDAMB-231 細(xì)胞的生長(zhǎng)有一定的抑制作用[9]。為了更全面地認(rèn)識(shí)和合理利用騰沖重樓,本研究對(duì)騰沖重樓進(jìn)行了化學(xué)成分研究和抗菌活性評(píng)價(jià)。從其根莖提取物中共分離出16 個(gè)化合物(圖1),分別鑒定為parisvanioside A(1)、kingianoside K(2)、7-氧薯蕷皂苷(7-oxodioscin,3)、pariposide A(4)、parisvanioside B(5)、parisrugoside H(6)、(25R)-17α-羥基螺甾-5-烯-3β-基O-β-D-吡喃葡萄糖基-(1→4)-O-[α-L-吡喃鼠李糖基-(1→2)]-β-D-吡喃葡萄糖苷((25R)-17α-hydroxyspirost-5-en-3β-ylO-β-D-glucopyranosyl-(1→4)-O-[α-L-rhamnopyranosyl-(1→2)]-β-Dglucopyranoside,7)、麥冬皂苷C?(ophiopogonin C?,8)、纖細(xì)薯蕷皂苷(gracillin,9)、重樓皂苷Ⅰ(paris saponin Ⅰ,10)、重樓皂苷II(paris saponin II,11)、重樓皂苷Ⅵ(paris saponin Ⅵ,12)、pennogenin 3-O-β-chacotrioside(13)、重樓皂苷H(paris saponin H,14)、重樓皂苷Ⅶ(paris saponin Ⅶ,15)和β-蛻皮激素(β-ecdysone,16)。其中化合物1~7 首次從騰沖重樓中分離得到。
HPD-100 型大孔樹脂(上海吉至生化科技有限公司),柱色譜用硅膠(80~100、200~300、300~400 目,青島美高集團(tuán)有限公司),反相(RP)C18硅膠(40~75 μm,F(xiàn)uji Silysia Chemical 有限公司),Sephadex LH-20 凝膠色譜(GE Healthcare Bio-Sciences AB),薄層色譜(TLC)板(100 mm×50 mm,青島美高化工有限公司),SK3300LH 型超聲波清洗器(上海科導(dǎo)超聲儀器有限公司),旋轉(zhuǎn)蒸發(fā)儀(瑞士BUCHI 公司),AL204 型電子分析天平(梅特勒托利多儀器有限公司),循環(huán)水真空泵(鞏義市予華儀器有限責(zé)任公司),SB-1300 型水浴鍋(上海愛朗儀器有限公司),500 MHz/800 MHz 核磁共振波譜儀(德國Brucker 公司),DFS 型快原子轟擊離子源質(zhì)譜儀(Thermo Fisher Scientific),Agilent 1200 Series 高效液相色譜儀半制備。
實(shí)驗(yàn)用騰沖重樓根莖樣品于2021 年10 月采自云南省蘭坪縣重樓種植基地,經(jīng)中國科學(xué)院昆明植物研究所楊珺實(shí)驗(yàn)師鑒定為藜蘆科重樓屬植物騰沖重樓P.tengchongensisY.H.Ji, C.J.Yang & Y.L.Huang,憑證標(biāo)本(YJ2021050)保存于中國科學(xué)院昆明植物研究所植物標(biāo)本室。
騰沖重樓干燥根莖(3 kg)粉碎,加70%乙醇(3L)60 ℃超聲提取6 次(每次1 h),濾過,取濾液,60 ℃減壓回收溶劑,得粗提物浸膏(660.0 g),加水(1 L)混懸,分別用等體積石油醚、醋酸乙酯和正丁醇分別萃取5 次,60 ℃減壓濃縮,得石油醚部分(118.1 mg)、醋酸乙酯部分(3.1 g)和正丁醇部分(182.0 g)。將正丁醇部分(170.2 g)用4 L 蒸餾水溶解,過 HPD100 大孔樹脂柱,乙醇-水(30%→50%→70%→95%)梯度洗脫后得4 部分:T1(1.7 g)、T2(5.4 g)、T3(22.7 g)和T4(50.0 g)。T1(1.7 g)經(jīng)RP C18硅膠柱色譜及Sephadex LH-20 凝膠柱(甲醇)分離純化后,得到化合物16(78.1 mg)。T2(5.4 g)經(jīng)RP C18硅膠柱色譜分段及HPLC(乙腈-水40∶60)分離純化后得到化合物15(4.9 mg,tR=21.4 min)和14(11.7 mg,tR=27.0 min)。T3(22.7 g)經(jīng)RP C18硅膠柱色譜得到T3-1(2.3 g)和T3-2(12.3 g)。T3-1 經(jīng)硅膠柱色譜(二氯甲烷-甲醇15∶1→1∶1)得到T3-1-1(57.9 mg)、T3-1-2(567.8 mg)和T3-1-3(34.7 mg)3 個(gè)組分。T3-1-1 用Sephadex LH-20 凝膠柱色譜(甲醇)及HPLC(甲醇-水77∶23)分離純化得到化合物1(1.1 mg,tR=25.5 min)、4(5.7 mg,tR=32.4 min)、7(2.8 mg,tR=22.3 min)和12(19.1 mg,tR=23.7 min)。T3-1-2用Sephadex LH-20凝膠柱色譜(甲醇)及HPLC(甲醇-水83∶17)分離純化得到化合物5(1.1 mg,tR=32.5 min)、6(1.1 mg,tR=23.0 min)、3(0.9 mg,tR=29.3 min)和13(26.9 mg,tR=25.7 min)。T3-1-3 經(jīng)HPLC(甲醇-水68∶32)分離純化后得到化合物2(0.5 mg,tR=34.2 min)。T3-2 經(jīng)硅膠柱色譜(二氯甲烷-甲醇15∶1→1∶1)得到T3-2-1(88.5 mg)、T3-2-2(1.0 g)和T3-2-3(10.1 mg)3 個(gè)組分。T3-2-1 用Sephadex LH-20 凝膠柱色譜(MeOH)及HPLC(甲醇-水85∶15)分離純化得到化合物8(3.5 mg,tR=25.0 min)。T3-2-2 經(jīng)RP C18硅膠柱色譜(80.7 mg)及HPLC(乙腈-水45∶55)分離純化得到化合物11(6.7 mg,tR=17.9 min)和9(5.2 mg,tR=21.6 min)。T3-2-3 經(jīng)反復(fù)重結(jié)晶(甲醇)得化合物10(9.1 mg)。

對(duì)分到的化合物進(jìn)行抗細(xì)菌和抗真菌活性實(shí)驗(yàn),參照文獻(xiàn)中的方法進(jìn)行活性評(píng)價(jià)[27]。
將單體化合物溶于DMSO 中,放入96 孔板中稀釋,在各孔中加入細(xì)菌菌液(大腸埃希氏菌ATCC25922、金黃色葡萄球菌金黃亞種ATCC29213、銅綠假單胞菌ATCC27853、耐甲氧西林金黃色葡萄球菌ATCC43300),終濃度為5×105CFU/mL;設(shè)置培養(yǎng)基為空白對(duì)照、細(xì)菌對(duì)照以及頭孢他啶、青霉素G 鈉陽性藥對(duì)照。37 ℃培養(yǎng)24 h,酶標(biāo)儀檢測(cè)625 nm 吸光度(A)值,計(jì)算細(xì)菌抑制率。

結(jié)果顯示,化合物1 和3~16 均對(duì)大腸埃希氏菌ATCC25922、金黃色葡萄球菌金黃亞種ATCC29213、銅綠假單胞菌ATCC27853、耐甲氧西林金黃色葡萄球菌ATCC43300 無明顯抑制作用,在化合物濃度為100 μmol/L 時(shí),其抑制率低于50%。
將化合物溶于DMSO 中,放入96 孔板中稀釋,在各孔中加入真菌菌液(絮狀表皮癬菌CBS 566.94、紅色毛癬菌ATCC4438、石膏樣小孢子菌CBS118893、白色念珠菌氟康唑耐藥株),終濃度為1×105CFU/mL(白色念珠菌氟康唑耐藥株)和5×105CFU/mL(絲狀真菌);設(shè)置培養(yǎng)基為空白對(duì)照、真菌對(duì)照以及兩性霉素B、鹽酸特比萘芬陽性藥對(duì)照。白色念珠菌氟康唑耐藥株37 ℃培養(yǎng)24 h,絲狀真菌25 ℃培養(yǎng)5 d,酶標(biāo)儀檢測(cè)625 nmA值,計(jì)算真菌抑制率。

當(dāng)濃度為100 μmol/L 時(shí),化合物3 對(duì)絮狀表皮癬菌和石膏樣小孢子菌抑制率為70%~74%;化合物5、6、8、9 對(duì)絮狀表皮癬菌、紅色毛癬菌和石膏樣小孢子菌的抑制率為65%~96%;化合物7、10~15 對(duì)白色念珠菌氟康唑耐藥株、絮狀表皮癬菌、紅色毛癬菌和石膏樣小孢子菌的抑制率為54%~100%(表1)。故進(jìn)一步對(duì)絮狀表皮癬菌、紅色毛癬菌和石膏樣小孢子菌有抑制活性的化合物進(jìn)行50%最低抑菌濃度(50% minimun inhibitory concentration,MIC50)測(cè)試,結(jié)果顯示,化合物3、5~15 對(duì)絮狀表皮癬菌有較強(qiáng)抑制作用(表2),MIC50為0.07~93.60 μmol/L;化合物5~15 對(duì)紅色毛癬菌有較強(qiáng)抑制作用(表3),MIC50為0.06~73.08 μmol/L;化合物3、5~8、10~15 對(duì)石膏樣小孢子菌有很強(qiáng)的抑制作用(表4),MIC50為0.04~69.92 μmol/L。

為了更進(jìn)一步發(fā)掘其利用價(jià)值,本研究對(duì)騰沖重樓正丁醇萃取物進(jìn)行系統(tǒng)的研究。通過對(duì)騰沖重樓根莖的提取、分離,共得到16 個(gè)化合物,其中化合物1~7 為首次從騰沖重樓中分離得到,豐富了騰沖重樓的化學(xué)成分類型。
前人僅報(bào)道了騰沖重樓中化合物的細(xì)胞毒活性[9],本研究較系統(tǒng)地評(píng)價(jià)了該物種中化合物的抗細(xì)菌和抗真菌活性。研究顯示滇重樓甲醇提取物可抑制宋內(nèi)氏痢疾桿菌、粘質(zhì)沙雷氏菌、大腸桿菌、金黃色葡萄球菌[28]。實(shí)驗(yàn)結(jié)果發(fā)現(xiàn)16 個(gè)化合物均對(duì)大腸埃希氏菌、金黃色葡萄球菌金黃亞種、銅綠假單胞菌、耐甲氧西林金黃色葡萄球菌無明顯抑制作用。皮膚癬菌是常見的臨床病原真菌,會(huì)引起人類頭發(fā)、皮膚及指趾甲的感染,可用兩性霉素B、鹽酸特比萘芬、伊曲康唑、伏立康唑、泊沙康唑、艾沙康唑等藥物進(jìn)行治療[29],但重樓皂苷進(jìn)行抗皮膚癬菌方面的研究較少,故進(jìn)行抗皮膚癬菌方面的研究。陸克喬等[30]的研究發(fā)現(xiàn),滇重樓的正丁醇提取物能有效抑制體外白色念珠菌的生物膜形成,可抑制白色念珠菌?;衔?、12~15 對(duì)白色念珠菌氟康唑耐藥株抑制率可達(dá)100%?;衔?1 對(duì)絮狀表皮癬菌、紅色毛癬菌、石膏樣小孢子菌抑菌效果最好,MIC50值分別為0.07、0.06、0.04 μmol/L。目前只對(duì)騰沖重樓進(jìn)行抗菌活性的研究,后續(xù)可進(jìn)行細(xì)胞毒活性等的測(cè)試,使對(duì)騰沖重樓的研究更加全面。騰沖重樓化合物對(duì)深部真菌白色念珠菌氟康唑耐藥株和3 種皮膚癬菌有較好的抑制作用,為今后對(duì)騰沖重樓治療皮膚及深部真菌感染疾病方面的開發(fā)利用提供參考。
