摘要 [目的]建立測定不同產(chǎn)地短柄五加根、莖中刺五加苷E含量的HPLC方法。[方法]采用Agilent TC-C 18(2) (250 mm×4.6 mm,5 μm)色譜柱;以乙腈-0.1%磷酸(16∶84)為流動相;流速1 mL/min;檢測波長220 nm,進樣體積為10 μL,柱溫30 ℃。[結(jié)果]刺五加苷E在8.0~160.6 μg/mL與峰面積呈良好的線性關(guān)系( r =0.999 6),平均加樣回收率為98.62%,RSD為0.50%。[結(jié)論]該方法操作簡便、穩(wěn)定可靠,適用于短柄五加根、莖中刺五加苷E含量的測定,可作為倒卵葉五加藥材的質(zhì)量控制指標。

Abstract [Objective] To establish an HPLC method for the simultaneous determination of eleutheroside E content in the roots and stems of ?Acanthopanax brachypus ??from different habitats.[Method] Separation was carried out on Agilent TC-C 18(2) column (250 mm×4.6 mm,5 μ m).The mobile phase consisted of acetonitrile -0.1% phosphoric acid water (16∶84) with a flow rate of 1.0 mL/min.The detection wavelengths was 220 nm, volume of injection was 10 μL, and the temperature of column was set at 30 °C.[Result]Eleutheroside E showed a good linear relationship between 8.0-160.6 μg/mL and the peak area ( r =0.999 6), the average recovery rate was 98.62%, and the RSD was 0.50%.[Conclusion]The method is convenient, stable and reliable, which is suitable for the determination of the content of eleutheroside E in the roots and stems of ?Acanthopanax brachypus ?Harms, and can be used as the quality control index of ?Acanthopanacis brachypus ?radix et rhizoma seu caulis.

倒卵葉五加 (Acanthopanacis brachypus ?radix et rhizoma seu caulis)為五加科植物短柄五加 Acanthopanax brachypus ?Harms [Eleutherococcus brachypus ?(Harms) Nakai]的干燥根和根莖或莖,分布于我國西北地區(qū)的陜西、甘肅、寧夏,主治體虛乏力、腰膝酸軟、脾腎陽虛、失眠多夢、食欲不振等病癥[1-2]。倒卵葉五加是陜西的特色植物藥,有很高的藥用及研究價值[3]?,F(xiàn)代藥理學研究表明,倒卵葉五加具有增強免疫力、抗疲勞、保護缺血心肌、抑制血小板聚集、抗血栓形成和抗氧化等藥理作用[4-10]。目前,對倒卵葉五加的研究主要集中在苷類和黃酮類化合物方面,對其具體化學成分的研究以及質(zhì)量控制的文獻比較少。倒卵葉五加藥材的質(zhì)量控制指標單一,主要以紫丁香苷的含量測定為標準[11-12]。近年來,采用HPLC法對刺五加、糙葉五加中刺五加苷E含量的測定均有研究報道[13-15],而倒卵葉五加中刺五加苷E的含量測定鮮見報道。通過該試驗的前期研究發(fā)現(xiàn),短柄五加根、莖中刺五加苷E的含量高于紫丁香苷。為了進一步有效控制倒卵葉五加的質(zhì)量,該研究選擇以刺五加苷E為檢測指標,試圖建立穩(wěn)定、高效的倒卵葉五加藥材中刺五加苷E的測定方法,并比較不同產(chǎn)地、不同部位的短柄五加中刺五加苷E含量的高低,以期為其資源的開發(fā)利用與藥材質(zhì)量控制提供科學依據(jù)。
