摘要:檢索Embase、Web of Science和PubMed,全面查找關(guān)于腫瘤類疾病風(fēng)險(xiǎn)與谷胱甘肽轉(zhuǎn)硫酶T1基因多態(tài)性關(guān)系的Meta分析的文獻(xiàn)。按疾病類型分別描述樣本量最大的Meta分析結(jié)果,以期明確二者關(guān)系。

谷胱甘肽轉(zhuǎn)硫酶T1(GSTT1)為解毒酶,其變異體是空白基因型,使其活性喪失,易導(dǎo)致多種腫瘤疾病。然而,由于疾病的種類不同,其與GSTT1基因多態(tài)性的關(guān)系也比較復(fù)雜。Meta分析可以提高統(tǒng)計(jì)效能,使不一致的原始研究合并得到最終可靠的結(jié)論。本研究檢索Embase、Web of Science和PubMed數(shù)據(jù)庫(kù),關(guān)于腫瘤類疾病風(fēng)險(xiǎn)與GSTT1基因多態(tài)性關(guān)系Meta分析的文獻(xiàn),綜合總結(jié)其結(jié)果。
1 資料與方法
本文檢索Embase、Web of Science和PubMed數(shù)據(jù)庫(kù)腫瘤類疾病風(fēng)險(xiǎn)與谷胱甘肽轉(zhuǎn)硫酶T1基因多態(tài)性關(guān)系的Meta分析的文獻(xiàn)。按疾病類型分別描述樣本量最大的Meta分析結(jié)果,以期明確二者關(guān)系。
2 結(jié)果
各種腫瘤最大樣本量的Meta分析結(jié)果,見表1。
3 討論
本研究總結(jié)出GSTT1空白基因升高了腫瘤類疾病患病風(fēng)險(xiǎn)的有:膀胱癌、前列腺癌、頭頸癌、鼻咽癌、乳腺癌、結(jié)直腸癌、胃癌、肝細(xì)胞癌、胰腺癌、急性髓細(xì)胞性白血病、慢性髓細(xì)胞性白血病、淋巴瘤、骨髓增生異常綜合征、肺癌、骨肉瘤;腫瘤類疾病患病風(fēng)險(xiǎn)與GSTT1空白基因無統(tǒng)計(jì)學(xué)意義相關(guān)的有:腎細(xì)胞癌、腦神經(jīng)膠質(zhì)瘤、腦膜瘤、喉癌、口腔癌、甲狀腺癌、宮頸癌、卵巢癌、結(jié)直腸腺瘤、食道癌、兒童急性淋巴細(xì)胞白血病、黑色素瘤。
各種腫瘤疾病與GSTT1空白基因多態(tài)性的不同結(jié)果,反應(yīng)了其易感性的不同,也受遺傳因素、環(huán)境因素等的影響。另外,Meta分析來源于各個(gè)不同的原始研究,可能存在臨床不一致性,包括人種、性別、年齡、基因多態(tài)性的測(cè)量方法、對(duì)照組來源等等。腫瘤發(fā)病的風(fēng)險(xiǎn)也不能單一從GSTT1基因多態(tài)性來解釋。盡管存在以上局限性,本文全面總結(jié)的腫瘤類疾病風(fēng)險(xiǎn)與谷胱甘肽轉(zhuǎn)硫酶T1基因多態(tài)性關(guān)系的Meta分析的文獻(xiàn)結(jié)果,為研究其發(fā)病風(fēng)險(xiǎn)提供了其中一條遺傳方面的線索。
