Breeding report of a new table Phyllanthus emblica cultivar Lütianyougan

Abstract: Phyllanthus emblica L. is one of important fruit crops with both medicinal and edible values. Currently, fresh variety of P. emblica has high economic benefit and great development potential. Herein, Lütianyougan, a new table variety of P. emblica with excellent traits, was selected from the wild seedling population. In 2013, a native population of P. emblica with different traits was first discovered in Hekou Town, Lühe county, Shanwei City. After many years of field investigation and quality assessment, the fruits from a single plant line was found, which was named as No. 2 tree, whose fruit was bigger than other varieties, and had strong aftertaste and light astringency. Due to its high quality and great potential in the market, No. 2 tree was grafted on wild seedling rootstocks for further field trials in 2014. Through 3 generations of continuous observation on the growth and fruiting habits of grafted seedlings, its offsprings showed stable economic characteristics and high-yield performance with good fruit quality, and its genetic stability was confirmed by inter simple sequence repeat (ISSR) analysis. During 2018—2021, the regional trials were carried out in three different cities including Shanwei, Jieyang and Shantou, and the growth and fruiting habits of No. 2 tree were stable with good performance. In November 2023, it was released as Lütianyougan with the registration code of Yuepingguo 20230009 after the validation by the Guangdong Provincial Crop Variety Examination and Approval Committee. Lütianyougan is a fresh-eating variety of P. emblica. The tree vigor is strong with erect shape. The color of bark is gray-brown, the old branches are offwhite with round-punctiform pores, while the new shoot is slender with dense longitudinal textures. The leaf is imparipinnate, young leaf is yellow-green, and mature leaf is dark green, leathery and rectangular in shape. The flowers are white with six petals. The inflorescence of Lütianyougan is cymes composed of many male flowers and one female flower or all male flowers. The fruit is oblate and smooth with visible ribs, the peel of immature fruit is blueish-green while the peel of mature fruit is yellowish-green, the pulp is light-green, crisp and slagging, with strong aftertaste, and its astringency is slight. The single fruit mass is 11.0 g, with the maximum of 17.34 g. The fruit shape index is 0.78 and the edible rate is 92.6%. The soluble solid content is 9.44%, the total sugar content is 7.2 g·100 g-1, the titrable acid content is 10.81 g·kg-1, and the vitamin C content is 294.0 mg·100 g-1. In Chaoshan area of Guangdong, the fruit ripens from early October to early November, belonging to the late-maturing variety type. The maturity and fruiting periods are long, and the physiological fruit drop is not obvious. It has large fruit, high yield, good marketability and strong adaptability. Due to high easiness to manage and cultivate, it is suitable for cultivation in Guangdong and similar climate areas.

余甘子(Phyllanthus emblica L.)是大戟科(Euphorbiaceae)葉下珠屬(Phyllanthus)一種熱帶、亞熱帶水果,其果實初食酸澀,良久乃甘,故名余甘子,又稱油甘子、牛甘果、滇橄欖、庵摩勒等[1]。余甘子果實含有豐富的維生素C、氨基酸、微量元素、有機(jī)酸、酚酸類、黃酮類、多糖類等營養(yǎng)活性物質(zhì)[2-3],為藥食同源,在中國栽培歷史悠久,自2000 a(年)前的東漢《異物志》到晉代《南方草本狀》、明朝《本草綱目》,再到現(xiàn)代的《中國藥典》均有收載,具有清熱利咽,潤肺化痰,健胃消食,收斂止瀉,提高免疫力、抗氧化、抗衰老、抗腫瘤、降血壓等功效,被聯(lián)合國衛(wèi)生組織指定為推廣種植的三種保健植物之一,具有良好的食用、保健和藥用價值[3-5]。除了干果做中藥材,余甘子在廣東、福建、廣西等地以鮮食為主,還可加工生產(chǎn)各類果脯、蜜餞、果茶、果汁、復(fù)合飲料、果醬、果酒、果粉、凍干片、袋泡茶等[3,6],具有較高的經(jīng)濟(jì)效益。
近年來,隨著余甘子果汁和茶飲風(fēng)靡全國,對鮮食余甘子的需求量越來越大[7]。廣東歷來有鮮食余甘子的習(xí)慣,是中國鮮食余甘子最大的消費市場和種植區(qū),也先后育成一批性狀優(yōu)良的地方品種(品系),包括玻璃油甘、白玉油甘、上湖仙油甘、珍珠油甘、柿餅油甘、獅頭油甘、矮種油甘等,有力支撐廣東余甘子產(chǎn)業(yè)的發(fā)展。酸澀味是影響余甘子鮮食品質(zhì)的重要因素之一[7],隨著市場的不斷變化和人們對高品質(zhì)鮮食余甘子的追求,一些酸澀味較重、口感差或果實小、產(chǎn)量低的地方品種已逐漸被淘汰,僅有少量種植。目前,廣東省余甘子產(chǎn)業(yè)面臨主栽品種單一、產(chǎn)期集中、酸澀味較重、口感不佳、適合鮮食和加工制汁的優(yōu)質(zhì)品種較少等問題[6],嚴(yán)重限制了余甘子產(chǎn)業(yè)的健康和高質(zhì)量發(fā)展。因此,亟須培育品質(zhì)優(yōu)良、豐產(chǎn)性好和適宜鮮食、制汁的余甘子新品種,以促進(jìn)品種結(jié)構(gòu)優(yōu)化和產(chǎn)業(yè)健康可持續(xù)發(fā)展。廣東的余甘子自然分布廣,野生種群類型多,蘊含豐富的變異類型,而且傳統(tǒng)多為實生苗繁殖,后代間存在不同的變異,單株中間性狀多,經(jīng)過長期的自然進(jìn)化和人工選擇形成了豐富的種質(zhì)資源。為了充分挖掘、利用廣東余甘子種質(zhì)資源,選育出適合鮮食和加工制汁的余甘子新品種,促進(jìn)廣東地區(qū)余甘子產(chǎn)業(yè)的健康發(fā)展,選育組開展了廣泛的資源調(diào)查、田間觀察和選種工作,經(jīng)過多年的努力,選育出鮮食余甘子新品種綠田油甘(圖1)。
1 選育過程
2013年,在汕尾市陸河縣河口鎮(zhèn)田墩村發(fā)現(xiàn)大片樹齡超過50 a(年)的野生油甘群體,經(jīng)過綜合鑒定分析,初步篩選出3株鮮食口感好的優(yōu)良單株,編號分別記為1~3號。對上述優(yōu)良單株,經(jīng)過連續(xù)多年田間調(diào)查和品質(zhì)分析,發(fā)現(xiàn)初選2號優(yōu)良單株系實生單株,該單株樹齡超過百年,長勢中等,分枝較多,樹形傘狀,結(jié)果性良好,果實個大、果皮光滑,果肉爽脆、酸甜適中,澀味輕,適合鮮食,且回甘濃,其果實大小和口感等與地方土種油甘明顯不同,符合筆者制定的選育目標(biāo),擬命名為綠田油甘,進(jìn)行系統(tǒng)性狀觀察調(diào)查。2014年從該2號優(yōu)良單株母樹采集枝條作為接穗,以地方土種油甘為砧木,在陸河縣河口鎮(zhèn)田墩村開展高接試驗。經(jīng)過田間觀察分析,2014年從母樹V0采集枝條作為接穗,發(fā)現(xiàn)其高接后代(V1)性狀和品質(zhì)保持不變;從2016年開始,又從V1后代采集枝條高接到土種油甘樹上(V2代),經(jīng)連續(xù)多年多代嫁接比較發(fā)現(xiàn),綠田油甘嫁接后代仍能保持母樹的優(yōu)良生物學(xué)性狀,表現(xiàn)性狀穩(wěn)定。2018—2022年在汕尾市、揭陽市、汕頭市等地進(jìn)行區(qū)域品種比較試驗,并對其果實的可溶性固形物、維生素C、可滴定酸、總糖含量等理化品質(zhì)指標(biāo)進(jìn)行檢測,連續(xù)多年田間觀察和品質(zhì)分析結(jié)果表明,綠田油甘生長勢強(qiáng),樹冠呈傘形,分枝力較強(qiáng),沒有明顯主干,適應(yīng)性較強(qiáng),其果實品質(zhì)優(yōu)良,口感好、澀味輕、回甘濃,商品性佳。2022年10月通過專家現(xiàn)場審查,2023年11月通過廣東省農(nóng)作物品種審定委員會審定,定名為綠田油甘(編號為粵評果20230009)。
2 品種分子鑒定
為判定綠田油甘是一個新的余甘子資源,選取綠田油甘、土種油甘、上湖仙油甘、矮種油甘、珍珠油甘、白玉油甘、獅頭油甘、玻璃油甘、柿餅油甘和六月白油甘等余甘子種質(zhì)的新生健康葉片,采用CTAB法提取基因組DNA,參照加拿大哥倫比亞大學(xué)UBC公布的96條引物序列,篩選出10條多態(tài)性好且條帶清晰的ISSR引物用于品種分子鑒定(引物序列見表1)。利用該10條ISSR引物對不同余甘子種質(zhì)DNA進(jìn)行PCR擴(kuò)增,共獲得725條清晰條帶,其中“841”引物的擴(kuò)增結(jié)果見圖2-A。利用NTSYS-pc2.1軟件計算遺傳相似系數(shù),并進(jìn)行相似性聚類分析。如圖2-C所示,綠田油甘(C4)及其母樹(C5)單獨聚為一類,與其他余甘子資源處于不同類群。
為測定綠田油甘嫁接后代的遺傳穩(wěn)定性,分別從母樹(V0)和嫁接后代(V1、V2和V3)提取DNA進(jìn)行PCR擴(kuò)增,兩條引物“890”和“891”的擴(kuò)增結(jié)果均表明,嫁接后代V1、V2和V3與母樹V0無差異條帶(圖2-B),能保持母樹的穩(wěn)定遺傳性狀。
3 主要性狀
3.1 植物學(xué)特征
綠田油甘樹姿開張,樹勢旺盛,樹冠呈圓傘形,疏密相間,主干不明顯,分枝能力強(qiáng)。樹皮灰褐色,老枝具有灰白色圓點狀皮孔;小枝條纖細(xì),具有致密的縱行紋理。幼年樹一年抽梢3~4次,成年結(jié)果樹一年抽發(fā)春梢和夏梢2次梢,春梢結(jié)果性較差,夏梢抽梢較短。2年生樹冠幅可達(dá)2.4 m×2.6 m,株高1.58 m,莖粗5.9 cm,一級分枝2~3個;3年生樹冠幅可達(dá)2.7 m×3.0 m,株高1.73 m,莖粗7.2 cm,一級分枝2~5個。奇數(shù)羽狀復(fù)葉,長12.3~25.8 cm;小葉數(shù)19~45枚,2列互生,嫩葉黃綠色,成熟葉片濃綠,革質(zhì),矩圓形,小葉片長1.66~2.81 cm,寬0.63~0.88 cm?;榘咨?,花瓣6瓣,聚傘狀花序由多朵雄花和1朵雌花或全為雄花組成,生于羽狀復(fù)葉中下部小葉葉腋。雌雄同株異花,雄花花梗長1.0~2.5 mm,3雄蕊,雄蕊黃色,花藥直立,長圓形,頂端具短尖頭,藥室平行,縱裂;雌花花梗長約0.5 mm,子房卵圓形,3室,3花柱,基部合生,頂端2裂,裂片頂端再2裂。果實扁圓形,果棱明顯,果腹線6條;果皮光滑,有光澤,未成熟時皮色青綠,成熟后果皮黃綠色;果肉淺綠色,肉質(zhì)爽脆,纖維少、化渣,澀味淡,回甘味濃。
3.2 果實主要經(jīng)濟(jì)性狀
綠田油甘果實平均單果質(zhì)量11.0 g,最大單果質(zhì)量達(dá)17.34 g;果實平均縱徑22.85 mm,平均橫徑29.30 mm,果形指數(shù)0.78,果核質(zhì)量0.82 g,可食率92.6%(表2)。果實扁圓形,果皮黃綠色,果面光滑;果肉淺綠色,肉質(zhì)爽脆、化渣(表3);可溶性固形物含量(w,后同)9.44%,總糖含量7.2 g·100 g-1,還原糖含量6.9 g·100 g-1,蔗糖含量0.28 g·100 g-1,可滴定酸含量10.81 g·kg-1,維生素C含量294.0 mg·100 g-1,粗纖維含量1.0%,單寧含量8.66 mg·g-1(表4)。與對照土種油甘相比,綠田油甘果實大,果肉厚,可食率高,果實形狀、大小和種皮顏色也明顯不同,并且可滴定酸、單寧和粗纖維含量均顯著降低,澀味輕,酸甜適中,口感好,回甘味濃,大小均勻,整齊度高,商品性好。
3.3 生物學(xué)特性
3.3.1" " "生長結(jié)果習(xí)性" " 綠田油甘樹勢健壯,老熟枝條為主要結(jié)果母枝,羽狀復(fù)葉可連續(xù)多年從1年生以上結(jié)果母枝同一萌發(fā)部位抽發(fā)出1~5個,落葉時整個脫落;聚傘花序伴隨春梢而萌發(fā),抽發(fā)能力強(qiáng),發(fā)于羽狀復(fù)葉中下部小葉葉腋;結(jié)果母枝連續(xù)結(jié)果能力強(qiáng),自然授粉坐果率高,在50%以上。嫁接苗第2年可開花結(jié)果,3年生樹和4年生樹每666.7 m2產(chǎn)量約為654.0 kg和1602.0 kg。
3.3.2" " 物候期" " 在廣東潮汕地區(qū),3月上中旬開始萌芽抽新梢,3月下旬至4月初始花,4月中旬盛花,4月下旬花謝,花期30 d左右;果實在10月上旬到11月上旬成熟,成熟期和掛果期較長。整個營養(yǎng)生育期240 d左右,為晚熟品種。與土種油甘相比,綠田油甘果實成熟期晚10~15 d。
3.3.3" " 適應(yīng)性和抗逆性" " 在汕尾、汕頭、揭陽等各試點的樹體生長健壯,樹勢旺盛,結(jié)果性好,穩(wěn)產(chǎn)性強(qiáng),且產(chǎn)量和果實品質(zhì)優(yōu)于土種油甘,表現(xiàn)為廣適性。在年降水量1200 mm以上、年平均氣溫21 ℃以上、極端最低氣溫大于1 ℃的區(qū)域種植可正常生長,喜偏酸性土壤,耐旱性強(qiáng),在山地丘陵和坡地均可正常生長,在排水性好的田地也可種植,結(jié)果性狀良好。
4 栽培技術(shù)要點
4.1 種植密度和建園技術(shù)
平地或緩坡地株行距宜3 m ×4 m至4 m ×5 m,山坡地株行距宜3 m ×4 m,每666.7 m2種植50~65株為宜。種植前挖好定植穴,施足基肥,每穴施腐熟農(nóng)家肥10.0~20.0 kg或商品有機(jī)肥5.0~10.0 kg、復(fù)合肥1.0 kg??赏ㄟ^定植嫁接苗或高接換種等途徑建園。
4.2 整形修剪
以培養(yǎng)低干、多主枝、分枝角度較大的開心形樹冠為主。2~3年生幼年樹生長時期,在主干高50~60 cm處剪頂定干,再在主干上培養(yǎng)3~4個主枝;待主枝長至約45 cm時剪頂,培養(yǎng)4~6個副主枝,選留枝條應(yīng)四面錯落伸展,分布合理。結(jié)果樹樹高應(yīng)控制在300 cm以下,樹冠有效結(jié)果母枝長度垂直方向控制在250 cm以內(nèi),水平方向控制在200 cm以內(nèi)。結(jié)果母枝以長枝、中枝為主,主要采用抹芽和疏枝等方式,避免短截。
4.3 肥水管理
4.3.1" " 幼年樹" " 定植成活后開始追肥,在春梢、夏梢和秋梢抽發(fā)前各施肥一次。植后第一年每株施尿素25.0 g和復(fù)合肥25.0~50.0 g,以后每年增加50%至1倍的施肥量。
4.3.2" " 結(jié)果樹" " 按平均單株產(chǎn)量50.0 kg計,每株年施豆餅或花生麩1.0~2.0 kg、有機(jī)肥5.0~7.5 kg、復(fù)合肥1.0~2.0 kg。每年施肥3次:促梢保果肥在2月中下旬春梢萌發(fā)前施用,以氮、鉀肥為主,配合磷、鈣肥;壯果促梢肥于6月中旬幼果開始膨大期施用,以鉀肥、磷肥為主,配施氮肥;采果復(fù)壯肥在采果后,以有機(jī)肥為主。
4.4 病蟲害防治
在廣東地區(qū),余甘子病蟲害相對較少,以防治銹病、炭疽病、介殼蟲和紅蜘蛛為主。應(yīng)貫徹“預(yù)防為主,綜合防治”的植保方針,加強(qiáng)農(nóng)業(yè)栽培管理,化學(xué)防治應(yīng)抓住病害初發(fā)期或害蟲低齡期用藥,減少農(nóng)藥使用量。
