摘要:采用雙水相萃取法輔助醇提法來提取純化杜仲(Eucommia ulmoides Oliver)中的總黃酮和綠原酸。通過單因素試驗(yàn)和Box-Behnken響應(yīng)面法確定最佳提取工藝,利用乙醇/無機(jī)鹽雙水相體系將杜仲提取物中的總黃酮和綠原酸進(jìn)行有效分離。結(jié)果表明,在最佳提取條件下,杜仲總黃酮的最佳雙水相萃取體系為乙醇的質(zhì)量分?jǐn)?shù)50%、K2HPO4的質(zhì)量分?jǐn)?shù)20%,此時(shí),相比R為2.93,分配比D為10.35,杜仲總黃酮的萃取率達(dá)77.94%。綠原酸的最佳雙水相萃取體系為乙醇的質(zhì)量分?jǐn)?shù)60%、K2HPO4的質(zhì)量分?jǐn)?shù)20%,此時(shí),相比R為3.43,分配比D為12.32,杜仲綠原酸的萃取率達(dá)78.22%。開發(fā)條件溫和、成相穩(wěn)定、用時(shí)較短的雙水相萃取技術(shù),對(duì)杜仲的開發(fā)利用有重要意義。

Abstract: In this study, the two-phase extraction method was used to assist the extraction of total flavonoids and chlorogenic acid in Eucommia ulmoides Oliver. The optimal extraction process was determined by single factor and Box-Behnken response surface method test, and the total flavonoids and chlorogenic acid in Eucommia ulmoides extract were separated by ethanol/inorganic salt aqueous two-phase system. The results showed that under the optimal extraction conditions, the best aqueous two-phase extraction system for total flavonoids in Eucommia ulmoides was 50% ethanol content with 20% mass fraction of K2HPO4, the phase ratio R was 2.93 and the distribution ratio D was 10.35, and the extraction ratio could reach 77.94%. The best aqueous two-phase extraction system for chlorogenic acid was 60% ethanol with 20% K2HPO4 in water, the phase ratio R was 3.43 and the distribution ratio D was 12.32 with the extraction ratio up to 78.22%. The dual aqueous phase extraction technology with mild development conditions, stable phase and short time is great significant for the development and utilization of Eucommia ulmoides.

杜仲(Eucommia ulmoides Oliver),落葉喬木,高可達(dá)20 m,樹皮灰褐色、粗糙,芽體卵圓形,外面發(fā)亮,紅褐色,葉橢圓形、卵形或矩圓形,薄革質(zhì),長6~15 cm,寬3.5~6.5 cm,在中國有著悠久的歷史,是中國獨(dú)有的一種珍稀樹種,被列為國家二級(jí)保護(hù)野生藥材物種。其性溫,味甘,歸肝、腎經(jīng),具有補(bǔ)肝腎、強(qiáng)筋骨、安胎的功效[1],在醫(yī)療、保健和食品領(lǐng)域,杜仲都有著良好的開拓空間。杜仲提取物主要活性成分有總黃酮和綠原酸,總黃酮具有降血壓、降血脂、抗氧化、抗腫瘤和調(diào)節(jié)骨代謝等廣泛的藥理作用[2-5],綠原酸具有降血壓、增高白血球、抗菌、抗病毒和腫瘤、增強(qiáng)免疫力、清除體內(nèi)自由基等功效[6-9]。因此,對(duì)杜仲總黃酮和綠原酸的提取工藝研究有著良好的發(fā)展前景。
目前,杜仲總黃酮和綠原酸常用的提取方法有水提法、酶解法和醇提取等。水提法提取液黏度大,過濾濃縮困難,提取物容易發(fā)生霉變;酶解法所需要的反應(yīng)時(shí)間長,所用的儀器與設(shè)備要求比較高,且酶是蛋白質(zhì),受溫度影響比較大,容易引起含糖成分的過濾困難;醇提法提取物成分復(fù)雜,有效成分分離難度較大,但提取率相對(duì)較高,如何將所需要的活性成分從其提取物中分離出來就顯得尤為重要。雙水相萃取技術(shù)是利用親水有機(jī)溶液的水溶液形成的雙水相,利用被分離物在兩相中溶解度的差異進(jìn)行兼具提取和分離雙重功能的高效分離技術(shù)[10,11]。利用雙水相體系可以為生物物質(zhì)提供良好的生物環(huán)境[12],可實(shí)現(xiàn)杜仲有效成分的提取分離[13,14]。
本研究以陜西省地道藥材杜仲為研究對(duì)象,采用雙水相萃取法輔助醇提法提取純化杜仲中的總黃酮和綠原酸。通過單因素試驗(yàn)和Box-Behnken響應(yīng)面法確定最佳提取工藝,再經(jīng)有機(jī)相與無機(jī)鹽形成的雙水相體系將杜仲中的總黃酮和綠原酸有效分離,運(yùn)用相平衡原理分析總黃酮和綠原酸在各類不同的無機(jī)鹽與有機(jī)相乙醇形成的雙水相體系中的分配性質(zhì)[15],實(shí)現(xiàn)杜仲總黃酮和綠原酸的分離與初步純化。
