摘要:晚脆蜜是六月鮮棗品種的控制雜交后代經(jīng)幼胚培養(yǎng)技術(shù)選育而成的鮮食棗新品種。該品種樹(shù)體矮化,樹(shù)冠緊湊,棗頭枝節(jié)間短,適宜密植栽培。果實(shí)長(zhǎng)橢圓形,果型較大,平均單果質(zhì)量為14.7 g。果實(shí)硬度大,肉質(zhì)致密,耐貯性較強(qiáng)。鮮棗可食率95.20%,可溶性固形物含量(w,后同)為28.4%,總糖含量27.5%,酸含量0.383%,維生素C含量3580 mg·kg-1。在山西太谷地區(qū),4月中旬開(kāi)始萌芽,6月上旬進(jìn)入盛花期,9月下旬果實(shí)成熟,果實(shí)生育期110d左右,屬于晚熟類型。早果豐產(chǎn)性能較強(qiáng),2~3年生枝的棗吊平均結(jié)果1.0個(gè),3~5年生樹(shù)產(chǎn)量6250kg·hm-2。果實(shí)抗裂果能力強(qiáng),裂果率僅為3.8%。該品種是一個(gè)樹(shù)體矮化、早果豐產(chǎn)、抗裂耐貯、晚熟優(yōu)質(zhì)的鮮食品種,具有良好的產(chǎn)業(yè)發(fā)展前景。

Breeding report of a new variety of Chinese jujube Wancuimi via embryo culture

Abstract:Ziziphus jujuba Mill.is one of the important fruit trees native to China.With the continuous changes in the market,fresh jujube variety has high economic benefit and great development potential.There is an urgent demand to breed new varieties of fresh jujube with both crack resistance and high fruit quality.The problem of jujube embryo abortion seriously restricts the process of jujube hybrid breeding.Embryo culture technology provides the possibility to solve the problem of early embryo abor-tion and offers a new way for breeding new variety.Breeders have attempted to use embryo culture to obtain jujube hybrid offspring,but so far there have been no reports on obtaining new varieties.Wancui-mi is a new cracking resistant and late maturing jujube variety obtained through immature embryo cul-ture technology after controlled hybridization of fresh jujube variety Liuyuexian with Mifengguan.The growth vigor of this variety was moderate and robust.The main trunk was longitudinally cracked,the branches were purple-red,the internodes were short,and the thorns were undeveloped.The leaf blade was ovate-lanceolate,slightly curled inward,with blunt apex,wedge-shaped base,and blunt teeth on the margin.The flowering amount was moderate,with diurnal opening.Compared with the maternal va-riety Liuyuexian,the internodes of this variety were shorter.This variety exhibited strong early yield performance,with an average of 1.0 fruit per hanging branch of 2-3 year-old branches.The graftedtrees began to bear fruit in the second year.The fruits of this variety were relatively large,oblong.The average single fruit weight was 14.7 g.The fruit shoulder was flat,the top was slightly protruding,and the fruit dots were small and dense.The flesh was dense,showing significantly higher fruit strength,flesh hardness,flesh thickness,and flesh compactness than Liuyuexian.The fruit color was red,with a sweet and sour taste,and good freshness.The edible rate of the fresh jujube was 95.2%,the soluble sol-id content was 28.4%,the soluble sugar content was 27.5%,the acidity was 0.383%,and the vitamin C content was 3580 mg·kg-1.The fruit core was middle-large,long-spindle-shaped,with plump kernels and a high kernel rate.The seeds were spindle-shaped and reddish-brown.This variety had strong crack resistance,with a low cracking rate of 3.8%,far lower than the cracking rate of 38.8%of the Li-uyuexian.In the Taigu county of Shanxi,bud burst was in mid-April,initial flowering in late May,peak flowering in early June,fruit coloring in mid-September,and fruit maturation in late September.The fruit development period was about 110 days,belonging to the late-maturing variety type.This variety was a fresh eating variety with compact tree body.

棗(Ziziphus jujuba Mill.)是原產(chǎn)中國(guó)的重要果樹(shù)之一。隨著市場(chǎng)的不斷變化,以制干棗品種為主的現(xiàn)狀難以滿足消費(fèi)者多樣化的需求,而且發(fā)展鮮食棗的經(jīng)濟(jì)效益高、發(fā)展?jié)摿Υ骩1]。但在生產(chǎn)中鮮食棗優(yōu)質(zhì)不抗裂或抗裂不優(yōu)質(zhì)的問(wèn)題較為突出,嚴(yán)重制約著鮮食棗產(chǎn)業(yè)的健康和高質(zhì)量發(fā)展。因此,亟須選育抗裂果兼具優(yōu)質(zhì)的鮮食棗新品種。
棗栽培歷史悠久,但育種技術(shù)進(jìn)展緩慢,長(zhǎng)期以來(lái)?xiàng)棙?shù)育種一直以芽變選種、株系選優(yōu)和實(shí)生選種為主,雜交育種十分困難[2]。主要原因是棗胚發(fā)育不良或中途敗育現(xiàn)象普遍存在,致使許多擁有優(yōu)良性狀的棗種質(zhì),因無(wú)法獲得后代而難以被利用,從而限制了親本的選擇,加大了雜交育種的局限性。胚培養(yǎng)技術(shù)為解決棗胚敗育問(wèn)題提供了可能,以期為加快品種選育進(jìn)程開(kāi)辟一條新途徑[3]。有研究者[4-8]試圖采用幼胚培養(yǎng)技術(shù)獲得棗雜種后代,但迄今未見(jiàn)獲得新品種的報(bào)道。筆者自2001年以來(lái)致力于棗胚培養(yǎng)技術(shù)研究,先后建立了棗胚培養(yǎng)體系[9]、幼胚經(jīng)愈傷組織培養(yǎng)的技術(shù)體系[10-12],成功獲得了一批胚培苗,為利用幼胚培養(yǎng)選育棗優(yōu)良品種提供了技術(shù)支撐。
六月鮮棗是山東省原產(chǎn)的中熟、質(zhì)優(yōu)的鮮食地方良種[13]。晚脆蜜棗是筆者于2010年開(kāi)始利用六月鮮為母本的控制雜交后代經(jīng)幼胚培養(yǎng)技術(shù)獲得,是國(guó)內(nèi)首次通過(guò)幼胚培養(yǎng)技術(shù)選育而成的鮮食良種,具有豐產(chǎn)、抗裂果、耐貯、優(yōu)質(zhì)、晚熟等特性。該品種的選育成功為棗育種技術(shù)和種質(zhì)創(chuàng)新開(kāi)辟了新途徑。
1選育過(guò)程
2010年5月在山西農(nóng)業(yè)大學(xué)(山西省農(nóng)業(yè)科學(xué)院)果樹(shù)研究所選取生長(zhǎng)發(fā)育正常的6株10年生六月鮮棗品種,在樹(shù)體上部各方位高接蜂蜜罐棗品種開(kāi)展控制雜交授粉試驗(yàn)。因六月鮮后代的種仁發(fā)育不飽滿,播種后很難獲得后代。而幼胚培養(yǎng)技術(shù)可以使許多具有特異性狀優(yōu)良棗種質(zhì)的幼胚得以挽救而獲得后代。2012年7月采集經(jīng)控制雜交后代的幼胚(胚齡30d,小球形胚階段),在無(wú)菌條件下進(jìn)行胚乳看護(hù)培養(yǎng),在培養(yǎng)基MS、水解乳蛋白0.5 mg·L-1、糖濃度7%,溫度(24±2)℃和光照(時(shí)間14 h·d-1,光照度3000 lx)條件下添加適宜的激素配比(0.5 mg·L-1 IBA+1.5 mg·L-1 ZT+3.0 mg·L-1 GA3+0.2 mg·L-1 NAA),成胚率達(dá)52.53%。幼胚經(jīng)胚乳看護(hù)培養(yǎng)后形成球狀體,將其完整剝離,放入2.0 mg·L-1 BA的培養(yǎng)基中進(jìn)行愈傷組織誘導(dǎo)。將誘導(dǎo)形成的愈傷組織轉(zhuǎn)接到添加0.5 mg·L-1 TDZ培養(yǎng)基中,在30 d后愈傷明顯增多且質(zhì)地良好,并有綠色芽點(diǎn)出現(xiàn),分化率達(dá)70.06%。將已獲得的胚培苗剪成留3~4片葉的莖段,接入MS+2.0 mg·L-1 BA+0.5 mg·L-1 IBA+0.06 mg·L-1 NAA的培養(yǎng)基上進(jìn)行擴(kuò)繁增殖。經(jīng)擴(kuò)繁后的胚培苗進(jìn)行生根培養(yǎng),生根率達(dá)90%。煉苗后,于2013年春移栽到大田栽培觀察。2016年從50個(gè)胚培養(yǎng)品系中篩選出編號(hào)為6-6的優(yōu)良品系。該品系不僅表現(xiàn)果實(shí)成熟期較晚、抗裂果、鮮食品質(zhì)優(yōu)異,而且還具有樹(shù)體生長(zhǎng)勢(shì)中庸、節(jié)間短等特性。2016年采集優(yōu)良品系6-6的接穗,經(jīng)嫁接繁殖、性狀觀察和區(qū)域試驗(yàn),表現(xiàn)性狀穩(wěn)定一致。經(jīng)與母本品種六月鮮比較,表現(xiàn)果實(shí)成熟期晚、果肉質(zhì)地致密及枝條節(jié)間短等特異性,經(jīng)SSR分子檢測(cè),DNA多態(tài)性高度相似,將其定名為晚脆蜜(圖1,圖2)。于2023年12月通過(guò)山西省林木品種審定委員會(huì)審定(良種編號(hào):晉S-SV-ZJ-014-2023)。
2主要性狀
2.1植物學(xué)特征
晚脆蜜樹(shù)體較小,樹(shù)姿較開(kāi)張。主干條狀皮裂,棗頭枝為紫紅色,節(jié)間短,節(jié)間長(zhǎng)度7.5 cm,二次枝節(jié)間長(zhǎng)度4.6 cm,彎曲度中等,針刺不發(fā)達(dá)。2~3年生枝每棗股棗吊數(shù)5.5個(gè),棗吊長(zhǎng)度22 cm,葉片數(shù)13枚。葉片中大,葉長(zhǎng)6.2 cm,寬3.1 cm,葉片卵狀披針形,微內(nèi)卷,葉尖鈍尖,葉基圓楔形,葉緣鈍齒?;恐械?,花序著花4~9朵,蜜盤乳黃色,花徑6.43 mm,晝開(kāi)型。該品種與六月鮮棗相比表現(xiàn)枝條節(jié)間較短(表1)。
2.2生長(zhǎng)結(jié)果習(xí)性
晚脆蜜樹(shù)體矮化,生長(zhǎng)勢(shì)中庸健壯,幼齡枝結(jié)果能力強(qiáng)。經(jīng)多年調(diào)查,果吊率較高,2~3年生枝的棗吊平均結(jié)果1.0個(gè),對(duì)照品種六月鮮為0.9個(gè)。嫁接樹(shù)第2年開(kāi)始結(jié)果,第3~5年平均產(chǎn)量6750kg·hm-2,早果豐產(chǎn)性能較強(qiáng)。晚脆蜜品種與對(duì)照品種六月鮮相比表現(xiàn)樹(shù)體生長(zhǎng)勢(shì)中庸健壯(表1)。
2.3物候期
在山西太谷地區(qū),晚脆蜜4月中旬開(kāi)始萌芽,5月下旬為初花期,6月上旬進(jìn)入盛花期,9月中旬果實(shí)開(kāi)始著色,9月下旬果實(shí)成熟進(jìn)入脆熟期。果實(shí)生育期110d左右,為晚熟品種類型。與六月鮮棗品種相比,成熟期晚10d以上(表2)。
2.4果實(shí)經(jīng)濟(jì)性狀
晚脆蜜果實(shí)較大,長(zhǎng)橢圓形,縱徑3.37 cm,橫徑2.56 cm,平均單果質(zhì)量14.7 g。果肩平,果頂稍突出,果點(diǎn)小而密。果實(shí)硬度大,肉質(zhì)致密。采用英國(guó)Stable Micro Systems公司生產(chǎn)的TA.XT plus質(zhì)構(gòu)儀測(cè)定,在果皮強(qiáng)度、果肉硬度、果肉粗細(xì)、果肉內(nèi)聚性等果肉質(zhì)地相關(guān)指標(biāo)顯著高于六月鮮。果實(shí)色澤紅色,風(fēng)味酸甜,鮮食口感好。鮮棗可食率95.2%,可溶性固形物含量28.4%,可溶性糖含量27.5%,酸含量0.383%,維生素C含量3580 mg·kg-1。果核中大,長(zhǎng)紡錘形,種仁飽滿,含仁率較高。種子倒紡錘形,紅褐色。晚脆蜜與對(duì)照品種六月鮮相比,果肉質(zhì)地表現(xiàn)致密(表3)。
2.5耐貯性、抗裂果性
晚脆蜜品種耐貯性較強(qiáng),在冷庫(kù)常規(guī)管理?xiàng)l件下,貯存2個(gè)月后,好果率仍可超過(guò)90%。該品種具有較強(qiáng)的抗裂果能力。裂果率較低,平均僅為3.8%,遠(yuǎn)低于六月鮮品種裂果率38.8%。
3栽培技術(shù)要點(diǎn)
3.1栽植密度和建園技術(shù)
適宜密植栽培,株行距(2.0~2.5)m×(3.0~4.0)m,每666.7 m2栽65~110株。根據(jù)立地條件、樹(shù)形及栽培模式等決定適宜的密度??赏ㄟ^(guò)定植嫁接苗、坐地酸棗苗嫁接和高接換優(yōu)等途徑建園。
3.2樹(shù)形和整形修剪技術(shù)
樹(shù)形采用小冠疏層形或紡錘形等。小冠疏層形樹(shù)高控制在1.8~2.0 m,干高30~50 cm,全樹(shù)有5~7個(gè)骨干枝,分二層著生在中央領(lǐng)導(dǎo)干上。紡錘形樹(shù)高控制在1.8~2.0 m,干高40~50 cm,自主干距離地面40 cm以上螺旋狀排列15~17個(gè)二次枝。該品種樹(shù)冠緊湊、生長(zhǎng)勢(shì)中庸健壯且枝條節(jié)間短,夏季修剪較為省工。夏剪主要采用抹芽、摘心、拉枝和環(huán)剝等措施,促進(jìn)早果豐產(chǎn)。冬剪以疏枝為主。
3.3花果管理技術(shù)
晚脆蜜具有較強(qiáng)的早果豐產(chǎn)性能,但花期采用環(huán)剝、環(huán)割、摘心、拉枝以及噴赤霉素、硼酸、磷鉀肥等措施可顯著提高產(chǎn)量。對(duì)設(shè)施栽培、肥水條件較好、樹(shù)勢(shì)偏旺的骨干枝在盛花期采取1次環(huán)割措施,可達(dá)到控勢(shì)增產(chǎn)的效果。該品種的結(jié)果能力強(qiáng),為實(shí)現(xiàn)其豐產(chǎn)穩(wěn)產(chǎn),除加強(qiáng)修剪、肥水管理外,還要注意合理負(fù)載。一般2~3年生的初果期樹(shù)應(yīng)控制在4500~5250 kg·hm-2,5年后盛果期不應(yīng)超過(guò)15 000 kg·hm-2。
3.4土肥水管理技術(shù)
適宜土層深厚、土壤肥力較高、有機(jī)質(zhì)含量高的壤砂土。萌芽期、棗頭枝旺長(zhǎng)期和果實(shí)膨大期對(duì)肥水需求較大,分別澆水1次,果實(shí)膨大期追施磷鉀肥300 kg·hm-2,秋季施有機(jī)肥45 000~75 000 kg·hm-2,并灌封凍水。
3.5病蟲(chóng)害防控技術(shù)
遵循“預(yù)防為主、綜合防治”的原則,注意綠盲蝽象、食芽象甲、棗銹病等病蟲(chóng)害的防治。
4市場(chǎng)前景展望
晚脆蜜是國(guó)內(nèi)首次通過(guò)棗幼胚培養(yǎng)技術(shù)選育而成的鮮食棗新品種,具有樹(shù)體矮化、早果豐產(chǎn)、抗裂果、耐貯、晚熟優(yōu)質(zhì)的特性。該品種解決了棗產(chǎn)業(yè)中抗裂果且優(yōu)質(zhì)鮮食棗品種稀缺的問(wèn)題,據(jù)該品種的特性、用途及各區(qū)試點(diǎn)的表現(xiàn),可在全國(guó)棗主要產(chǎn)區(qū)發(fā)展推廣,在國(guó)內(nèi)市場(chǎng)上有巨大的潛力和廣闊的前景。
