斷奶日齡對(duì)肉兔腸道發(fā)育的影響

摘要:本試驗(yàn)旨在研究斷奶日齡對(duì)肉兔腸道發(fā)育的影響。試驗(yàn)選擇初生肉兔120窩,隨機(jī)分成4組,每組30窩,分別在出生后21、25、28和35日齡斷奶,試驗(yàn)期56 d。結(jié)果表明:1)斷奶日齡對(duì)肉兔28和35日齡胃、小腸和盲腸重有顯著或極顯著影響(P<0.05或P<0.01),隨著斷奶日齡延后呈增加趨勢(shì),斷奶日齡對(duì)49和56日齡各消化器官重和各日齡小腸長(zhǎng)度無(wú)顯著影響(P>0.05);2)斷奶日齡對(duì)十二指腸(28、35和49日齡)、空腸(28、35、42和49日齡)和回腸絨毛高度(28、35、42和49日齡)有顯著或極顯著影響(P<0.05或P<0.01),隨著斷奶日齡延后呈先增加后降低趨勢(shì),而對(duì)56日齡各腸段絨毛高度無(wú)顯著影響(P>0.05),肉兔斷奶后絨毛高度需要2~3周才能恢復(fù),21日齡斷奶組恢復(fù)最慢,35日齡斷奶組恢復(fù)最快;3)斷奶日齡對(duì)十二指腸(28和35日齡)、空腸(28、35和42日齡)和回腸隱窩深度(35日齡)有顯著影響(P<0.05),隨著斷奶日齡延后呈降低趨勢(shì),而對(duì)49和56日齡各腸段隱窩深度無(wú)顯著影響(P>0.05);4)斷奶日齡對(duì)28和35日齡十二指腸、空腸和回腸絨毛高度/隱窩深度有極顯著影響(P<0.01),隨著斷奶日齡延后呈增加趨勢(shì),而對(duì)56日齡各腸段空腸絨毛高度/隱窩深度無(wú)顯著影響(P>0.05)。由此可見(jiàn),斷奶日齡對(duì)肉兔腸道49日齡前發(fā)育影響較大,對(duì)49日齡后影響變?。辉缙跀嗄探档土诵∧c黏膜絨毛高度,增加了隱窩深度;隨斷奶日齡延后,斷奶造成的肉兔小腸黏膜受損程度減輕,斷奶后一般需要2~3周才能恢復(fù)小腸黏膜結(jié)構(gòu)。

現(xiàn)代集約化養(yǎng)兔要求充分發(fā)揮母兔的繁殖潛能,縮短胎次間隔,提高母兔年產(chǎn)仔數(shù),對(duì)仔兔進(jìn)行早期斷奶是發(fā)揮母兔繁殖潛能的重要措施之一[1]。斷奶對(duì)仔兔是一個(gè)巨大的應(yīng)激,由于仔兔胃腸道發(fā)育不完善,容易發(fā)生消化道疾病,造成腹瀉死亡[2]。因此,研究掌握不同斷奶日齡對(duì)肉兔胃腸道發(fā)育和功能影響的變化規(guī)律,對(duì)指導(dǎo)肉兔科學(xué)早期斷奶具有重要意義。1978年,Chen等[3]和Rao等[4]研究了斷奶日齡(28、42、56日齡)對(duì)肉兔生產(chǎn)性能的影響,結(jié)果發(fā)現(xiàn)不同斷奶日齡對(duì)體重、飼料效率和胴體品質(zhì)無(wú)影響,死亡率隨斷奶日齡延后而降低,認(rèn)為28日齡斷奶最為經(jīng)濟(jì)。De Bias等[5]研究發(fā)現(xiàn)斷奶日齡(25和35日齡)顯著或極顯著影響干物質(zhì)和消化能的攝入量、49及63日齡重,但是對(duì)平均日增重或77日齡重?zé)o顯著影響,從而認(rèn)為25日齡斷奶更佳。Ferguson等[6]發(fā)現(xiàn)14日齡斷奶,并于14~21日齡進(jìn)行人工喂奶和補(bǔ)飼的兔與28日齡斷奶的對(duì)照組兔相比,70日齡體重低0.2 kg,仔兔數(shù)和采食量差異不顯著,表明人工喂奶技術(shù)是可行的。在關(guān)于早期斷奶的研究中,不同斷奶日齡條件下商品肉兔在不同時(shí)間胃腸道發(fā)育的差異研究的報(bào)道很少。因此,亟需深入研究不同斷奶日齡對(duì)肉兔胃腸道形態(tài)和功能影響的變化規(guī)律,為肉兔早期斷奶技術(shù)的實(shí)現(xiàn)提供理論參考。本試驗(yàn)旨在研究不同斷奶日齡對(duì)肉兔消化道形態(tài)發(fā)育和功能的影響,為最佳斷奶日齡的確定和飼料配方研制提供科學(xué)依據(jù)。
1材料與方法
1.1試驗(yàn)設(shè)計(jì)
選擇初生新西蘭兔120窩,隨機(jī)分為4組,組間仔兔數(shù)和窩重差異不顯著(P>0.05),每組30窩,分別在21、25、28和35日齡斷奶,分別記為W21、W25、W28以及W35組,試驗(yàn)期為56 d。
1.2飼養(yǎng)管理
試驗(yàn)?zāi)竿糜蓪?zhuān)人管理,飼養(yǎng)管理一致,產(chǎn)仔當(dāng)日通過(guò)寄養(yǎng)的方式使每只母兔均帶養(yǎng)8只仔兔,哺乳期采用母仔分離飼養(yǎng),仔兔每日09:00哺乳1次,喂奶時(shí)將仔兔放入母兔籠中,哺乳10 min左右。16日齡時(shí)仔兔開(kāi)始補(bǔ)飼,自由采食飼喂。所有兔只均在35日齡當(dāng)天打耳號(hào),并根據(jù)體型大小和性別進(jìn)行分群分籠飼養(yǎng),60 cm×60 cm籠位中飼喂3只仔兔。42日齡時(shí)注射兔瘟-巴氏桿菌二聯(lián)疫苗1.5 mL/只。小兔一直飼喂肉兔飼糧直至試驗(yàn)結(jié)束,飼糧組成及營(yíng)養(yǎng)水平見(jiàn)表1。所有兔只自動(dòng)飲水,兔舍每天清潔1次。
1.3指標(biāo)測(cè)定及方法
每組分別在21、28、35、42和56日齡選擇體重接近群體平均值的6只兔,公母各占1/2,于當(dāng)日早上飼喂后1 h進(jìn)行屠宰,頸靜脈放血處死后,立即打開(kāi)腹腔,結(jié)扎幽門(mén)瓣、回盲瓣,將消化道取出,按照解剖學(xué)特征小心剝離,將小腸按解剖特征把十二指腸、空腸和回腸分別結(jié)扎。
1.3.1消化道重量和長(zhǎng)度測(cè)定
將小腸腸袢與腸道小心剝離,用軟尺測(cè)定其自然長(zhǎng)度即為小腸長(zhǎng)度。將胃、小腸和盲腸清洗掉內(nèi)容物,并在濾紙上去掉多余的水分,分別稱取重量,即為消化道重量。取樣部分估算后計(jì)入總重。
1.3.2小腸黏膜形態(tài)的測(cè)定
分別截取十二指腸、空腸和回腸中段2 cm組織,用生理鹽水沖洗后迅速放入預(yù)先配制好的10%福爾馬林溶液中固定。按常規(guī)方法制作石蠟切片,蘇木精-伊紅染色,按照Sun等[7]的方法測(cè)定絨毛高度和隱窩深度,計(jì)算絨毛高度/隱窩深度(V/C)。從絨毛頂端到陷窩頂端測(cè)定絨毛高度,從相鄰絨毛的內(nèi)陷部分測(cè)定隱窩深度。

1)預(yù)混料為每千克飼糧提供 The premix provided the following per kg the diet:Fe 100 mg,Cu 20 mg,Zn 90 mg,Mn 30 mg,Mg 150 mg,VA 4 000 IU,VD31 000 IU,VE 50 mg,膽堿 choline 1 mg。
2)消化能為計(jì)算值,其余為實(shí)測(cè)值。DE was a calculated value, while other nutrient levels were measured values.
1.4統(tǒng)計(jì)方法
試驗(yàn)數(shù)據(jù)用Excel 2007軟件進(jìn)行處理后,采用SPSS 17.0統(tǒng)計(jì)軟件進(jìn)行單因素方差分析,Duncan氏法進(jìn)行多重比較,以P<0.05為差異顯著性判斷標(biāo)準(zhǔn),結(jié)果用“平均值±標(biāo)準(zhǔn)差”表示。
2結(jié)果與分析
2.1斷奶日齡對(duì)消化器官重量和小腸長(zhǎng)度的影響
由表2可知,總體上,隨斷奶日齡延后,胃、小腸、盲腸重均呈增加趨勢(shì)。斷奶日齡顯著影響28、35以及42日齡的胃重(P<0.05),而對(duì)21、49和56日齡的胃重影響不顯著(P>0.05);斷奶日齡極顯著影響28日齡小腸重(P<0.01),顯著影響35日齡小腸重(P<0.05),而對(duì)42、49和56日齡的影響不顯著(P>0.05);28和42日齡的盲腸重組間存在顯著差異(P<0.05),35日齡的盲腸重組間存在極顯著差異(P<0.01),其中28、35和42日齡時(shí),早期斷奶的W21組低于其他各組,但是差異隨斷奶日齡延后逐漸減小,49日齡后各組差異不顯著(P>0.05)。斷奶日齡對(duì)小腸長(zhǎng)度無(wú)顯著影響(P>0.05)。

2.2斷奶日齡對(duì)小腸黏膜絨毛高度的影響
由表3可知,總體上,隨斷奶日齡延后,小腸黏膜絨毛高度呈先增加后降低趨勢(shì)。斷奶日齡顯著或極顯著影響肉兔28、35和49日齡十二指腸絨毛高度(P<0.05或P<0.01),斷奶日齡對(duì)42和56日齡影響不顯著(P>0.05);斷奶日齡顯著或極顯著影響28、35、42和49日齡空腸、回場(chǎng)絨毛高度(P<0.05或P<0.01),對(duì)56日齡影響不顯著(P>0.05)。可見(jiàn),隨日齡增加,斷奶日齡對(duì)小腸絨毛高度影響逐漸下降,斷奶后2~3周內(nèi)小腸絨毛得到恢復(fù)。

2.3斷奶日齡對(duì)小腸黏膜隱窩深度的影響
由表4可知,總體上,隨著斷奶日齡延后,小腸黏膜隱窩深度呈降低趨勢(shì)。斷奶日齡顯著影響28和35日齡十二指腸隱窩深度(P<0.05),對(duì)42、49和56日齡隱窩深度無(wú)顯著影響(P>0.05);斷奶日齡顯著影響28、35和42日齡的空腸隱窩深度(P<0.05),對(duì)49和56日齡隱窩深度無(wú)顯著影響
(P>0.05);斷奶日齡對(duì)35日齡回腸隱窩深度的影響顯著(P<0.05),對(duì)28、42、49和56日齡隱窩深度無(wú)顯著影響(P>0.05)。可見(jiàn),斷奶日齡對(duì)小腸前段黏膜隱窩深度的影響要大于后段的影響,斷奶后1~2周內(nèi)小腸黏膜隱窩深度得到恢復(fù),恢復(fù)時(shí)間短于絨毛高度。

2.4斷奶日齡對(duì)小腸黏膜V/C的影響
由表5可知,總體上,隨著斷奶日齡延后,小腸黏膜V/C呈增加趨勢(shì)。斷奶日齡極顯著影響十二指腸28和35日齡的V/C(P<0.01),顯著影響49日齡的V/C(P<0.05),對(duì)42和56日齡無(wú)顯著影響(P>0.05);斷奶日齡極顯著影響空腸28、35和42日齡的V/C(P<0.01),對(duì)49和56日齡無(wú)顯著影響(P>0.05);斷奶日齡極顯著影響回腸28和35日齡的V/C(P<0.01),對(duì)42、49和56日齡無(wú)顯著影響(P>0.05)??梢?jiàn),斷奶對(duì)肉兔小腸黏膜V/C的影響主要集中在斷奶后1~2周內(nèi),斷奶越早影響越大。

3討論
3.1斷奶日齡對(duì)消化器官重量和小腸長(zhǎng)度的影響
本研究發(fā)現(xiàn),斷奶日齡對(duì)28和35日齡的胃、小腸和盲腸重有著顯著或極顯著影響,斷奶越早,同日齡消化器官越小,斷奶越晚同日齡消化器官越大,49日齡后不同斷奶日齡組消化器官重差異不顯著,可見(jiàn),肉兔存在補(bǔ)償發(fā)育。Xiccato等[8]研究發(fā)現(xiàn)18日齡斷奶仔兔與30日齡斷奶仔兔35日齡體重差異較大,但是50日齡體重差異不大,該研究也驗(yàn)證了本試驗(yàn)結(jié)果,肉兔存在補(bǔ)償發(fā)育生長(zhǎng),斷奶日齡對(duì)于肉兔出欄階段的體重不存在顯著影響。本試驗(yàn)發(fā)現(xiàn),斷奶日齡對(duì)小腸長(zhǎng)度無(wú)顯著影響,可能與小腸的局部生長(zhǎng)有關(guān),肉兔小腸發(fā)育可能優(yōu)先發(fā)育長(zhǎng)度,后發(fā)育黏膜結(jié)構(gòu)。試驗(yàn)結(jié)果顯示,肉兔腸道發(fā)育過(guò)程存在補(bǔ)償生長(zhǎng),這為早期斷奶提供了可能。
3.2斷奶日齡對(duì)肉兔小腸黏膜發(fā)育的影響
本試驗(yàn)中,斷奶日齡對(duì)28、35和42日齡肉兔小腸絨毛高度、隱窩深度和V/C有較大影響,斷奶越早腸道黏膜絨毛高度越低,隱窩深度越深。斷奶過(guò)晚腸道黏膜絨毛高度也減少,隱窩深度也增加,但是變化幅度較小。顧憲紅等[9]研究不同斷奶日齡仔豬腸道發(fā)育規(guī)律時(shí)發(fā)現(xiàn)仔豬斷奶越早,仔豬產(chǎn)腸黏膜受損越嚴(yán)重,恢復(fù)時(shí)間也越長(zhǎng),一般斷奶后8~12 d降至最低,隨后恢復(fù)較快,仔豬和仔兔斷奶后腸道黏膜發(fā)育類(lèi)似。Bivolarski等[10]研究也發(fā)現(xiàn),在整個(gè)肉兔生長(zhǎng)階段,早期斷奶和正常斷奶兔的平均絨毛高度差異不顯著,但是在斷奶后第1天組間的絨毛高度和隱窩深度差異顯著。本試驗(yàn)同時(shí)也發(fā)現(xiàn),斷奶日齡對(duì)56日齡腸道黏膜結(jié)構(gòu)無(wú)顯著影響,說(shuō)明肉兔在斷奶后經(jīng)過(guò)2~3周時(shí)間后,腸道黏膜都得到恢復(fù),但早期斷奶組肉兔腸道黏膜恢復(fù)時(shí)間較長(zhǎng)。
4結(jié)論
① 斷奶日齡對(duì)肉兔腸道49日齡前發(fā)育影響較大,對(duì)49日齡后影響變小。
② 早期斷奶降低了小腸黏膜絨毛高度,增加了隱窩深度。
③ 隨斷奶日齡延后,斷奶造成的肉兔小腸黏膜受損程度減輕,斷奶后一般需要2~3周才能恢復(fù)小腸黏膜結(jié)構(gòu)。

Effects of Weaner Days of Age on Intestinal Development of Meat Rabbits

Abstract:This experiment was conducted to evaluate the effects of weaner days of age on intestinal development of meat rabbits. A total of 120 nests of new born meat rabbits borned were selected and randomly divided into four groups with 30 nests per group. The meat rabbits of four groups were weaned by the age of 21, 25, 28 and 35 days, respectively. The experiment lasted 56 days. The results showed as follows: 1) weaner days of age had significant effects on the weight of stomach, small intestine and caecum of rabbits by the age of 28 and 35 days (P<0.05), which tended to be increased by delayed weaner days of age, however, the weight of digestive organs and the length of small intestine of rabbits by the age of 49 and 56 days were not significantly affected (P>0.05). 2) Weaner days of age had significant effect on the height of villus of duodenum (28, 35 and 49 days of age), jejunum (28, 35, 42 and 49 days of age) and ileum (28, 35, 42 and 49 days of age) (P<0.05 or P<0.01), which tended to be firstly increased and then decreased by delayed weaner days of age, however, weaner days of age had no significant effect on the height of villus of duodenum, jejunum and ileum by the age of 56 days (P>0.05); the height of villus took 2 to 3 weeks to recover after weaned, the group weaned by the age of 21 days recovered slowest, and the group weaned by the age of 35 days recovered fastest. 3) Weaner days of age had significant effect on the depth of crypt of duodenum (28 and 35 days of age), jejunum (28, 35 and 42 days of age) and ileum (35 days of age) (P<0.05), which tended to be decreased by delayed weaner days of age, however, weaner days of age had no significant effect on the the depth of crypt of small inrestine of rabbits by the age of 49 and 56 days (P>0.05). 4) Weaner days of age had significant effect on villus height/crypt depth (V/C) of duodenum, jejunum and ileum of rabbits by the age of 28, 35 days (P<0.01), which tended to be increased by delayed weaner days of age, however, weaner days of age had no significant effect on V/C of small intestine of rabbits by the age of 56 days (P>0.05). It is concluded that weaner days of age can affect the intestinal development of rabbits before 49 days of age, but has little influence on that of rabbits after 49 days of age; early weaning can reduce the height of intestinal mucosal villus, but increase the depth of crypt; of earlier-weaned group; the damage on mucosa of small intestinal of rabbits can be alleviated by the delayed weaner days of age, and it generally require 2 to 3 weeks for rabbit to restore the structure of mucosa of small intestine.[Chinese Journal of Animal Nutrition, 2016, 28(1):102-108]
