3結(jié)論
集落實(shí)驗(yàn)和RTPCR實(shí)驗(yàn)結(jié)果顯示:在實(shí)驗(yàn)所采用的顆粒濃度范圍內(nèi),F(xiàn)e3O4納米顆粒對(duì)HeLa細(xì)胞基本沒有細(xì)胞毒性.對(duì)于核苷酸修復(fù)通路有缺陷的XPF細(xì)胞,F(xiàn)e3O4納米顆??梢砸种圃摷?xì)胞的生長(zhǎng)和分裂,表現(xiàn)出一定的細(xì)胞毒性.Fe3O4納米顆??梢允筙PF細(xì)胞內(nèi)CHEK1,RPA1,RPA2,RPA3基因轉(zhuǎn)錄比較明顯地增加.CHEK1基因轉(zhuǎn)錄的增加可以使XPF細(xì)胞的細(xì)胞周期停止,從而抑制其分裂和生長(zhǎng).而RPA1,RPA2,RPA3基因轉(zhuǎn)錄增加暗示了Fe3O4納米顆粒對(duì)XPF細(xì)胞產(chǎn)生某種損害.但究竟是何種損害需要進(jìn)一步研究.本文的研究結(jié)果表明:Fe3O4納米顆粒在腫瘤診斷、治療上的應(yīng)用必須謹(jǐn)慎,對(duì)有先天核苷酸剪切修復(fù)通路突變的腫瘤患者,就不能使用Fe3O4納米顆粒,否則會(huì)對(duì)患者造成額外的損害.
