摘要:目的 建立甘草渣中黃酮提取物的質(zhì)量標(biāo)準(zhǔn),為甘草渣中提取的總黃酮提供質(zhì)量控制的方法及依據(jù)。方法 采用薄層色譜法將提取物與甘草苷及甘草對照藥材對比鑒別,采用紫外分光光度法測定總黃酮含量,EDTA二鈉滴定法測定鈣離子含量。結(jié)果 供試品色譜中,在與對照藥材色譜相應(yīng)的位置上顯相同顏色的斑點,在與對照品相應(yīng)的位置上顯相同顏色的熒光斑點。紫外分光光度法測定甘草苷在3.32~9.96 ?g/mL范圍內(nèi)線性關(guān)系良好(r=0.999 7),吸光度在0.3~0.7之間,平均加樣回收率為102.41%(RSD=3.22%),3批甘草渣黃酮提取物中總黃酮平均含量為8.82%,鈣離子平均含量為0.107%(RSD=3.61%)。結(jié)論 本方法重復(fù)性好、靈敏度高、結(jié)果準(zhǔn)確,可用于甘草渣中黃酮提取物的質(zhì)量控制。

Abstract: Objective To establish the quality standard of flavone extracts in licorice residue; To provide the methods and basis for the quality control of total flavonoids extracted from licorice residue. Methods TLC was applied to compare the extracts with liquiritin and licorice; UV-spectrophotometry was used to measure the content of total flavones; EDTA-2Na titration was used to determine the content of calciumion. Results In the chromatogram of the test product, the spots of the same color were displayed on the corresponding position of the control medicinal material, and the fluorescent spots of the same color were displayed on the corresponding position of the control product. UV-spectrophotometry showed glycyrrhizin had a good linear relationship in the range of 3.32–9.96 ?g/mL (r=0.999 7), the absorbance between 0.3 and 0.7. The average recovery rate was 102.41% (RSD=3.22%), and the average content of total flavonoids in three batches of flavonoids extract was 8.82%. The average content of calciumion was 0.107% (RSD=3.61%). Conclusion This method has good repeatability, high sensitivity and accurate results, and can be used for the quality control of flavonoids in licorice residue.

甘草為豆科甘草屬甘草Glycyrrhiza uralensis Fisch、脹果甘草Glycyrrhiza inflata Bat.、光果甘草Glycyrrhiza glabra L.的干燥根和根莖,有效成分復(fù)雜,主要為甘草酸和黃酮類化合物[1],但目前開發(fā)、生產(chǎn)的主要是甘草中的甘草酸[2],剩余物作為藥渣棄去。研究表明,甘草渣中富含黃酮類化合物[3],具有多種生物活性,除消炎、抗菌、抗變態(tài)反應(yīng)等,在抗氧化、抗癌、防癌、防腫瘤及保肝護(hù)消化道等方面作用顯著[4]。
近年來,隨著對甘草藥理作用的進(jìn)一步認(rèn)識,全球?qū)Ω什莸男枨笕找嬖鲩L,野生甘草資源急劇減少,與此同時,甘草提取甘草酸或甘草浸膏后剩余的殘渣(甘草渣)也逐漸成為危害生態(tài)環(huán)境的又一物質(zhì)。本課題前期進(jìn)行了“基于化學(xué)轉(zhuǎn)化法破壁富集甘草藥渣中總黃酮的開發(fā)研究”(蘭州市科技局,2014-2-32)。為了保證甘草渣黃酮提取物的質(zhì)量穩(wěn)定性,本研究參考2015年版《中華人民共和國藥典》(以下簡稱《中國藥典》)甘草項進(jìn)行甘草渣黃酮提取物的研究,建立其薄層色譜鑒別法、總黃酮含量測定法及鈣離子含量測定方法等質(zhì)量標(biāo)準(zhǔn),以期為甘草渣中黃酮資源的有效、合理開發(fā)和質(zhì)量評價提供依據(jù)。endprint
